Objective:Esophageal Carcinoma(ESCA)is a malignant tumor of the alimentary canal with high incidence and mortality in our country.In many studies,DTL has been found to be a potential oncogene of tumor and an important regulatory factor of tumor cell proliferation and metastasis.Studies have found that DTL is highly expressed in ESCA tumor tissues,but its biological significance in ESCA has not been further verified and analyzed.Therefore,by analyzing the expression level of DTL in ESCA and its influence on tumor development and prognosis,this study explored whether DTL gene could be used as a therapeutic and prognostic marker for ESCA.Methods:Firstly,ONCOMINE and TIMER2 databases analyzed the transcriptome data of all tumors in TCGA to explore the mRNA expression level of DTL in tumor tissue,while GEPIA database was used to verify the expression results of DTL in ESCA.The UALCAN online site was used to continue the analysis of the relationship between DTL mRNA expression and clinicopathological features of ESCA.The R package of "Cluster Profiler" and STRING database preliminarily explored the action pathway and target genes of DTL in ESCA.Meanwhile,the "GSVA" R package was used to analyze the relationship between DTL expression and tumor immune infiltrating cells in TCGA-ESCA dataset.The "p RRophetic" R package performs drug sensitivity analysis to identify potential treatments for ESCA associated with DTL.Finally,the tumor tissue and clinicopathological data of ESCA were collected,and the DTL protein expression levels in 95 tumor tissue samples and adjacent normal tissue samples were detected by immunohistochemical method.The collected clinicopathological data were sorted out,and correlation analysis,Kaplan-Meier survival analysis and COX regression analysis were conducted with the protein expression level of DTL in ESCA to reveal the relationship between DTL and prognosis survival of ESCA patients.Results:Online database results showed that mRNA expression levels of DTL were elevated in a variety of tumor tissues,including ESCA,compared to normal tissues.UALCAN analysis of the relationship between DTL and ESCA clinicopathological factors found that there was a positive correlation between DTL mRNA expression and age,smoking level,drinking level and stage of patients.The results of enrichment analysis and immunoinfiltration analysis showed that the mechanism of DTL action on ESCA was mainly concentrated in the process of cell proliferation and metastasis,and the expression of DTL mRNA was negatively correlated with the infiltration levels of neutrophils,Th17 cells and B cells.The results of drug sensitivity analysis showed that several macromolecular compounds,such as palbocinb,rebocillin and abesilil,are expected to be potential therapeutic drugs for ESCA.The results of protein level verification were consistent with the trend of mRNA expression.The protein expression level of DTL in ESCA tissues was significantly higher than that in neighboring normal tissues,and DTL expression was correlated with tumor number and histological grade.The results of prognosis and survival analysis showed that upregulation of DTL can shorten the overall survival of ESCA patients,and may be an independent risk factor for poor prognosis in ESCA patients.Conclusions:DTL is highly expressed in ESCA tumor tissues,and its high expression significantly affects the prognosis and immune infiltration level of ESCA patients.It may be involved in the regulation of the proliferation of ESCA tumor cells,and is expected to become a potential target for molecular therapy and prognosis evaluation of ESCA patients.
