| Objective:Hepatocellular carcinoma(HCC)is the main type of liver cancer and is one of the most common malignant tumors.In recent years,there has been significant development in the treatment of HCC,but the insidious onset and rapid progression of HCC makes early diagnosis difficult,which directly contributes to the poor prognosis of HCC patients.A number of studies have shown that Denticleless E3 ubiquitin protein ligase homolog(DTL)is important in tumor development,progression and metastasis.This paper investigates the correlation between the expression level of DTL in HCC and its prognosis,clinicopathological features and immune infiltration,with the aim of providing new strategies to improve the diagnosis and treatment of HCC.Methods:1.To study the expression levels of DTL mRNA in a variety of tumors through the Oncomine database.2.To study the relationship between DTL mRNA expression levels and overall survival of HCC patients through the GEPIA database.3.To study the co-expressed genes of DTL through LinkedOmics database and perform GO functional annotation and KEGG pathway analysis.4.To study the relationship between DTL and immune infiltration of HCC was investigated by TIMER database.5.To analyse the relationship between DTL protein expression levels and clinicopathological features of HCC patients by immunohistochemical staining experiments and to explore the prognostic risk factors.Results:1.The mRNA expression levels of DTL were higher in a variety of tumors than in control tissues.2.Patients with high DTL mRNA expression had lower OS than those with low expression.3.GO functional annotation and KEGG pathway analysis of DTL co-expressed genes showed that DTL co-expressed genes were enriched in cell cycle,spliceosome and other pathways.The biological processes included DNA replication,chromosome segregation,RNA location.The cellular components included splicesomal complex,chromosomal region,nuclear chromatin,microtubule.The biological functions included catalytic activity,acting on DNA,histone binding,ATPase activity,tubulin protein binding.4.DTL mRNA expression levels were positively correlated with tumour cell purity,B cells,CD8+T cells,CD4+T cells,macrophages,neutrophils and dendritic cells in the immune microenvironment.In addition,DTL positively correlated with a variety of immune markers.5.Immunohistochemical staining experiments showed that the protein-positive expression of DTL in HCC tissues was lower than that in control tissues and correlated with tumour diameter and envelope integrity,K-M survival analysis showed that patients with low DTL expression had a worse prognosis and COX risk analysis showed that DTL expression and cirrhotic nodules were not only predictors of poor prognosis in HCC,but DTL expression and cirrhotic nodules were independent risk factors for poor prognosis of HCC.Conclusion:The expression level of DTL correlates to some extent with the prognosis,clinicopathological features and immune infiltration of HCC and has potential as a biomarker and immune-related therapeutic target for HCC. |
