Objective:Interstitial cystitis(IC)/Bladder Pain Syndrome(BPS)is a class of chronic diseases severely reducing quality of life,and its diagnosis is still a thorny problem.NLRP3(NOD-like Receptor Thermal Protein Domain Associated Protein 3)Inflammosomes play an important role in a variety of inflammatory diseases.The objective of this study is to explore the role of NLRP3 inflammatories in the occurrence and development of IC/BPS and to find relevant diagnostic markers.Methods:Three IC/BPS datasets(GSE11839,GSE11783,and GSE57560)were collected from the Gene Expression Omnibus database(GEO).Then merge them and debatch effects.The NLRP3 inflammasome-related gene came from the Msigdb database.Differentially expressed NLRP3 inflammasome-associated genes(NLRP3-DEGs)were analyzed between normal and IC/BPS tissues,followed by GO and KEGG enrichment assays to assess the function of NLRP3-DEGs.Then,the single sample Gene Set Enrichment Analysis(ss GSEA)method was used to explore the immune invasion landscape in IC/BPS and the association between NLRP3-DEGs and IC/BPS immune infiltration was analyzed.Machine learning methods were then used to screen for diagnostically valuable genes from NLRP-DEGs,and a nomogram model was created to assist diagnosis.Finaly,predict the gene-drug targets of NLRP3-DEGs through the DGIdb database.Results:A total of 12 NLRP3-DEGs were identified,including NLRC3,PSTPIP1,PLCG2,PTPN22,BTK,CARD8,AIM2,HSP90AB1,HMOX1,GBP5,PYCARD,TLR6 and others.Most NLRP3-DEGs were up-regulated in IC/BPS samples.2 signature genes were identified as NLRC3 and PSTPIP1 by machine learning algorithms,and the resulting diagnostic model was constructed and showed good performance.Immune infiltration analysis revealed that most immune cells differed between normal and IC/BPS tissues and that there was a strong correlation between NLRP3-DEGs and most immune cell scores and functions.In addition,potential drugs for the treatment of IC/BPS have been explored.Conclusion:NLRP3 inflammasome-related genes may be involved in the development of IC/BPS and regulate immune response.NLRC3 and PSTPIP1 have some diagnostic value for IC/BPS.These findings provide new directions for the diagnosis and treatment of IC/BPS and help clinical decision.Predicted drugs provide direction for the exploration of new IC/BPS treatments.
