Objective:To investigate the effects of NLRP3 inflammasome activator(LPS+ATP)and inhibitor(MCC950)on the proliferation,apoptosis and expression levels of NLRP3 inflammasome mRNA and its downstream cytokines IL-1β and IL-18 in MDS cell line MUTZ-1.Methods:The MUTZ-1 cell line was cultured and divided into three groups:control group(PBS group),activation group(LPS+ATP group),and inhibition group(MCC950+LPS+ATP group).The cell proliferation rate was detected by CCK-8 method,apoptosis rate by flow cytometry,cytokine IL-1β and IL-18 expression levels by ELISA,and NLRP3mRNA expression level by real-time fluorescence quantitative PCR(RT-qPCR).Results:Compared with the control group,the activation group significantly promoted MUTZ-1 cell proliferation(P<0.05),had a tendency to inhibit apoptosis(P>0.05),and the expression levels of NLRP3 inflammatory vesicle mRNA and its downstream cytokines IL-1β and IL-18 were elevated(P<0.05).And further addition of inhibitor significantly inhibited the effect of activator on MUTZ-1 cells(P<0.05).Conclusions:Intervention of the MUTZ-1 cell line with NLRP3 inflammasome activators and inhibitors affects cell proliferation,apoptosis and the expression of NLRP3 inflammasome mRNA and its downstream cytokines IL-1β and IL-18,indicating that NLRP3 inflammasomes play an important role in the MUTZ-1 cell line.
