| Coronary heart disease(CHD),a disease with the highest prevalence of cardiovascular diseases,has obvious genetic tendency and is affected by a variety of complex factors such as hyperlipidemia,hypertension,diabetes,smoking history and inflammatory factors.Among them,dyslipidemia is the most common risk factor for CHD and plays a leading role in the development of systemic atherosclerosis.The expression level of vascular scavenger receptor(CD163)can reflect the severity of atherosclerosis,which is expected to be an index to evaluate the CHD.Statins are the first-line drugs to treat hyperlipidemia.However,the therapy of atorvastatin has shown high individual variability.Therefore,this study proposes the hypothesis that single nucleotide polymorphisms of rs4883263 and rs11054072,which can affect the function of CD163 gene,may be related to the blood lipid level of patients with coronary heart disease.It can affect the process of CHD by affecting the blood lipid level of patients,whica may directly or indirectly affect the therapeutic effect of lipid lowering drug intervention.A total of 187 peripheral blood samples of inpatients and physical examination population(116 males,71 females)from December 2016 to May2020 were divided into CHD group(n=97)and control group(n=90).The differences between CHD group and control group were analyzed with all indexes,and the risk factors of CHD also were analyzed.Statistical methods were employed for comparing the differences of genotype distribution between CHD patients and control group so as to analyze the correlation between the polymorphisms of the site on various indicators of blood lipid in two groups.And monitoring of blood lipid levels in patients with coronary heart disease of different genotypes after taking atorvastatin calcium tablets.The correlation between the risk factors of CHD and the polymorphisms of the above-mentioned gene loci,the blood lipid level and the efficacy of statins were analyzed.The main findings of this research are as follows:1.CHD was closely related to the level of blood lipid,and the levels of triglyceride and apolipoprotein A were significantly increased in CHD group.In addition,the sex ratio(male/female)in the CHD group was 69/28,which was a significant difference.Therefore,CHD may have a male gender bias.Multivariate Logistic regression analysis showed that uric acid,apolipoprotein A 1 and low levels of high-density lipoprotein choleterol were also risk factors of CHD.2.The main genotype of CD163 rs4883263 was homozygous CC,and the main allele was C.The main genotype of CD163 rs11054072 was AG,and the main allele was A.And there was statistically different between CHD group and control group in the genotype distribution.After sex stratification,the genotype distribution of male CHD group was different from that of male control group,which suggesting that men with A allele may have a higher risk of CHD.3.This study found for the first time that CD163 rs4883263 and rs11054072 polymorphisms were related to blood lipid levels.Statistics results display that the TT genotype at rs4883263 may be associated with high levels of low-density lipoprotein choleterol.The AA genotype at rs11054072 may be related to the high total cholesterolăLDL-C and HDL-C levels.4.This study also confirmed that the levels of TC,TG,LDL-C in patients with CC and CT genotype at CD163 rs4883263 decreased significantly after treatment(p<0.05).The levels of TG in patients with TT genotype decreased after treatment(p<0.05).After treatment,the levels of TC,TG,LDL-C and apo B in patients with AA genotype at CD163 rs11054072 decreased significantly.The levels of TG and LDL-C in patients with AG genotype decreased significantly,and the levels of LDL-C and apo B in patients with GG genotype decreased significantly(p<0.05).The changes of blood lipids in CHD patients with different genotypes after treatment indicated that genotypes may lead to diffierences in the lipid-lowering effect of atorvastatin calcium tablets. |
