| ObjectiveCoronary heart disease,as the most common cardiovascular disease in clinic, has a high risk of cardiovascular events despite active treatment.Diabetes mellitus,as an equal risk of coronary heart disease, significantly increases the incidence and mortality of cardiovascular events. It has been proved that DPP-4 gene polymorphism is closely related to the occurrence of cardiovascular events. In this paper, the polymorphism of DPP-4 gene and its relationship with blood lipid and the degree of coronary artery stenosis were studied for the first time in patients with coronary heart disease complicated with type 2 diabetes mellitus.MethodsFrom January 2016 to June 2018,168 patients with coronary heart disease and type 2diabetes mellitus were collected in the Department of Cardiology,Qianfoshan Hospital,Shandong Province. The polymorphism distribution of rs3788979,an important SNP locus associated with blood lipid and cardiovascular disease, has been detected in the literature. The general information of each patient, including hospitalization number, sex, age, height, weight,BMI,blood glucose control, smoking history, drinking history and family history, etc were collected. At the same time, the Hb A1 c,FPG,TG,TC,HDL-C,LDL-C,LPa,Apo A1,apo B and HCY were extracted. The distribution of DPP-4 gene polymorphism and its relationship with blood lipid level as well as the degree of coronary artery stenosis were studied by univariate and multivariate analysis.ResultsThe distribution of SNP rs3788979 was polymorphic in the 168 patients with coronary heart disease and type 2 diabetes mellitus, which was in accordance with Hardy-Weinberg equilibrium(χ2=0.004,P =0.95).There were three different genotypes of rs3788979,AA,AG and GG.AA was defined as wild type, while AG and GG was mutant type.Compared with wild type patients, there was significant difference between the serum Apo B level in mutant patients(P=0.029).No significant difference was discovered in serum TG(P=0.833),TC(P=0.791),HDL-C(P=0.810),LDL-C(P=0.475),Lpa(P=0.774),Apo A1(P=0.370)and HCY(P=0.623).There was no significant difference in the severity of coronary artery stenosis among different genotypes in the 168 patients(P=0.185>0.05).The total samples were further divided into smoking group and non-smoking group. The results showed that there was no significant difference in coronary artery stenosis among different genotypes in smoking group(P=0.329>0.05),but in non-smoking group, signifycant difference was identified(P=0.027<0.05).ConclusionIn this study, the coronary heart disease patients complicated with type 2 diabetes mellitus was selected as the target population for the first time.It was found that the distribution of DPP-4 gene was polymorphic in the 168 patients. The polymorphism was associated with the level of blood lipid and the severity of coronary artery disease. If verified,it will be of great significance for the identification of high risk patients and the prediction of cardiovascular events, and thus bring new targets for the development of biomarkers and drugs, which may eventually reduce the incidence of cardiovascular events. |
