| Purpose:Non-Alcoholic Fatty Liver Disease(NAFLD)is the most common cause of chronic liver disease and an important cause of hepatocellular carcinoma.The occurrence of NAFLD is related to genetic,environmental,metabolic and other factors,mainly manifested as the accumulation of triglycerides in liver cells.The protein encoded by the PNPLA3 gene has triglyceride hydrolase activity,which can promote triglyceride hydrolysis.Although there have been a large number of studies on the polymorphism of PNPLA3 rs738409 and other loci,there is no study on the relationship between PNPLA3rs3747207 loci and NAFLD in China.This study aimed to investigate the correlation between PNPLA3 rs3747207 polymorphism and the risk of NAFLD in Qingdao Han population.Methods:(1)A total of 223 patients with NAFLD who attended Qingdao Municipal Hospital from December 2020 to June 2022 were stochastically enrolled in NAFLD group;168 healthy individuals were stochastically enrolled in control group.(2)We collected the clinical date and calculate body mass index(BMI).Blood sample of all subjects were taken to detect relevant biochemical parameters,including total cholesterol(TC)、low-density lipoprotein(LDL)、alkaline phosphatase(ALP)、high-density lipoprotein(HDL)、triglycerides(TG)、alanine aminotransferase(ALT)、 Gama-glutamyl transferase(GGT)、aspartate aminotransferase(AST)、total bilirubin(TBil)、fasting plasma glucose(FPG).(3)Polymerase chain reaction(PCR)and Matrix-assisted laser desorption/ionization time-offlight mass spectrometry(MALDI-TOF MS)determined the genotype of PNPLA3rs3747207.(4)The clinical data and biochemical indexes were analyzed by SPSS 26.0software,and P < 0.05 was considered as statistically significant meaning.The chi-square test was applied to compare qualitative data between groups.After the test of homogeneity of variance,if the two groups of continuous variables were normal,t-test was used for intergroup comparison,otherwise non-parametric test was used for inter-group comparison.The binary logistic regression analysis was used to investigate the risk factors for NAFLD.Results:(1)Comparing the clinical data and biochemical examination between the NAFLD and healthy control groups,there were significant differences in age,BMI,FPG,ALT,AST,ALP and TG between NAFLD group and healthy control group(all P < 0.05),However,there were no significant differences in gender,TC,LDL and TBil between the two groups.(all P > 0.05)(2)PNPLA3 rs3747207 had AA、AG and GG genotypes by sequencing.Analysis showed that there was no significant difference in allele and genotype distribution of rs3747207 between NAFLD group and control group(all P > 0.05).(3)Binary logistic regression analysis showed that GG genotype carriers didn’t increase the susceptibility of NAFLD(OR = 0.702,95%CI: 0.428-1.152,P = 0.161).After adjusting for age,gender and BMI,GG genotype carriers still didn’t increase the risk of NAFLD(OR= 0.593,95%CI: 0.332-1.062,P = 0.079).(4)In all subjects,the GG genotype carriers had higher LDL level than the non-carriers(P <0.05).(5)In the healthy control group,the GG genotype carriers had higher FPG、TC and LDL levels than the non-carriers(all P < 0.05).Conclusions:(1)In the Han population in Qingdao area,PNPLA3 rs3747207 allele and genotype distribution was not significantly associated with the susceptibility of NAFLD.(2)PNPLA3 rs3747207 GG genotype carriers was associated with higher levels of FPG,TC and LDL. |
