Objective: To explore the anti-inflammatory and analgesic effects of chitosan oligosaccharide and Antarctic Krill peptide on urate(MSU)crystal induced mouse gout model and the regulatory effects on hyperuricemia metabolic syndrome.Methods: 1.Effects of chitosan oligosaccharide and Antarctic Krill peptide on gouty arthritisThe mice were randomly divided into normal control group,model control group,Chitosan oligosaccharide prophylactic administration group(1.6 g/kg,0.8 g/kg,0.4g/kg),chitosan oligosaccharide therapeutic administration group(1.6 g/kg),Antarctic krill peptide prophylactic administration group(1.0 g/kg,0.5 g/kg,0.25g/kg),Antarctic krill peptide therapeutic administration group(1.0 g/kg)and positive drug group,prophylactic administration for 14 days.A mouse model of gouty arthritis induced by MSU crystals was established on the 8th day of prophylactic administration,and the treatment group was given medication on the day of the establishment of the gouty arthritis model for 7 consecutive days.To observe the effects of chitosan oligosaccharide and Antarctic krill peptide on the swelling degree of foot pad,mechanical pain and thermal pain in mice.2.Effects of chitosan oligosaccharide and Antarctic Krill peptide on inflammatory response in mouse balloon modelsMice were randomly divided into normal control group,model control group,chitosan oligosaccharide administration group(1.6 g/kg,0.8 g/kg,0.4 g/kg),Antarctic krill peptide administration group(1.0 g/kg,0.5 g/kg,0.25 g/kg)and positive drug group.The balloon model of mice was established on the 8th day of drug intervention.On the 14 th day,mouse gout balloon model was induced by MSU crystal.6 h later,the contents of IL-1β and IL-6 and the number of inflammatory cells were detected by balloon lavage solution,and the synovium was collected for pathological examination.3.Effects of chitosan oligosaccharide and Antarctic Krill peptide on components of hyperuricemia metabolic syndromeMice with spontaneous hyperuric acid(Uox knockout)were randomly divided into hyperuric acid model group,chitosan oligosaccharide administration group(1.6g/kg)and krill peptide administration group(1.0 g/kg).WT mice were also used as control.The administration group was treated with chitosan oligosaccharide and Antarctic krill peptide for 6 weeks,and the serum biochemical indexes and other components of metabolic syndrome were detected.Results: 1.A mouse model of gouty arthritis was established,and the swelling degree of the foot pad in the model group was significantly increased,and the mechanical pain threshold at the swollen part of the foot pad was significantly decreased.After the intervention of chitosan oligosaccharide and Antarctic krill peptide,the degree of foot swelling was significantly improved in the low-dose group of chitosan oligosaccharide,and the mechanical pain threshold in the swelling of the foot pad was significantly increased in the groups of chitosan oligosaccharide and Antarctic krill peptide high-dose group.2.A gout balloon model was established in mice.In the model group,the number of inflammatory cells,inflammatory factors IL-1β and IL-6 increased significantly in the balloon lavage solution,and a large number of inflammatory cells penetrated the superficial layer of the balloon synovium.The number of inflammatory cells and the content of IL-6 decreased significantly after the intervention of chitosan oligosaccharide and krill notholeptide,but there was no significant improvement on IL-1β.The infiltration of inflammatory cells in balloon synovium was significantly reduced in chitosan oligosaccharide groups and in Antarctic krill peptide high-dose and medium-dose groups.3.Serum uric acid,serum creatinine,urea nitrogen and cholesterol levels were significantly increased in mice with uric acid oxidase gene knockout.The levels of creatinine,urea nitrogen,cholesterol and triglyceride in blood of mice with spontaneous high uric acid decreased significantly after 6 weeks of chitosan oligosaccharide and Antarctic krill peptide intervention.Conclusions: Chitosan oligosaccharide and Antarctic krill peptide can not only significantly relieve joint swelling and mechanical pain induced by MSU crystal in gout joint inflammation,improve inflammatory cell infiltration in synovium tissue of gout balloon model,reduce the content of inflammatory factor IL-6 in balloon lavage solution,but also significantly reduce the levels of blood creatinine,urea nitrogen,cholesterol and triglyceride in mice with spontaneous hyperuric acid.The results indicated that chitosan oligosaccharide and Antarctic krill peptide had certain curative effect on gout inflammation and hyperuricemia metabolic syndrome.
