Eupolyphaga sinensis Walker is a true genus insect of the family trichosanthes,and its female dried body is the traditional Chinese medicine of China.At present,most of the turtle breeding in China has become a scale,but the lack of standardized production standard operating technical regulations,resulting in the quality of turtle insects mixed.In order to improve the quality and reduce the economic loss,it is urgent to establish the standard operating procedure for the breeding of Eupolyphaga Steleophaga,so as to standardize and standardize the breeding of Eupolyphaga Steleophaga.As a result,this paper intends to explore the effects of different feeding methods,drying temperatures and storage methods on the quality of the medicinal materials of E.sinensis Walker.(1)Treat the to-be-harvested E.sinensis Walker for 3 days and 7 days with fasting without substrate,fasting with substrate,and feeding with substrate.Observe the distribution of food residues in the digestive tract of the E.sinensis Walker,determine its growth indicators,intrinsic quality,and aflatoxin content.the results showed that after fasting,the E.sinensis Walker emptied the food residues in the digestive tract for 3 days without substrate and 7 days with substrate;after 3 days of treatment,there was no significant difference in the survival rate of the E.sinensis Walker in each group,but the protein,polysaccharide,and crude fat content of the E.sinensis Walker in the fasting group were significantly reduced(P<0.05).The total ash,acid-insoluble ash,and aflatoxin of the E.sinensis Walker in the fasting group without matrix the content was significantly lower than the other two groups,and the extract content,anticoagulant and fibrinolytic activity were significantly higher than the other two groups(P<0.05).After 7 days of treatment,the survival rate and specific growth rate of the E.sinensis Walker in the fasting group were significantly reduced(P<0.05),and the content of the extract,antithrombin and fibrinolytic activity of the E.sinensis Walker in the fasting group without matrix were significantly lower than that of the 3 days treatment(P<0.05).3 days before harvesting,the E.sinensis Walker were fasted without substrates to increase the extract content without affecting the yield,reduce the total ash and aflatoxin content,and at the same time increase the anticoagulant and fibrinolytic activities,and enhance the quality of the medicinal materials..(2)Treat the to-be-harvested E.sinensis Walker with the best feeding method selected in(1),and dry them with hot air at 60℃,70℃,80℃,90℃,100℃to constant weight,and measure their dryness.parameters,intrinsic quality and aflatoxin content.the results showed that as the drying temperature increased,the protein content first increased and then decreased,the 70℃drying group was significantly higher than the other groups(P<0.05);the crude fat content decreased,and the 60℃drying group was significantly higher than the other groups(P<0.05);the extract content decreased,and the 60 and 70℃drying group was significantly higher than the other groups(P<0.05).The anticoagulant activity and cellolytic activity increased first and then decreased.The70℃dry group was significantly higher than the other groups(P<0.05).The content of aflatoxin in the 60℃dry group was significantly higher than that of the other groups(P<0.05).there were no significant differences in polysaccharide content,total ash content and acid-insoluble ash content among the groups.In summary,70℃hot-air constant temperature drying is a more suitable processing method for E.sinensis Walker.It can not only reduce the crude fat and aflatoxin content of medicinal materials,but also increase the extract content,anticoagulant activity and cellolytic activity.(3)Use(1)and(2)to screen the best feeding method and drying temperature for the to-be-harvested E.sinensis Walker,pack them in four materials:ziplock bags,woven bags,cartons,and envelopes,and place them at 25°C,65RH.After being stored for 3 months,the intrinsic quality and aflatoxin content were measured after removal and compared with the medicinal materials before storage(YP).The results showed that the polysaccharide and crude fat content of the Eupolyphaga Steleophaga in the envelope group were lower than the other groups,and the protein content was significantly lower than that of the other groups(P<0.05);the content of the extract,anticoagulant activity and fibrinolytic activity of the Eupolyphaga Steleophaga in the ziplock bag group were significantly higher In other groups(P<0.05),the aflatoxin content was significantly lower than other groups(P<0.05).In summary,ziplock bags are relatively suitable packaging materials for the storage of Eupolyphaga Steleophaga.During storage for 3 months,moisture fluctuations are small,no mildew will occur,and the production of aflatoxins can be effectively controlled.At the same time,the extract content of medicinal materials is high and anticoagulant.Strong activity and cellolytic activity.(4)Use the best feeding method and drying temperature selected by(1)and(2)to make the water content of the pharmacopoeia upper limit 10%(Y)and constant weight 3%(H)of the E.sinensis Walker to be harvested,and place them at low temperature and low humidity(4°C,60%RH),medium temperature and medium humidity(25°C,65%RH),high temperature and high humidity(40°C,80%RH)environment,serial number is(Y4、H4)、(Y25、H25)and(Y40、H40),after 3 months of storage,the medicinal materials were taken out to determine the internal quality and aflatoxin content of the medicinal materials of the Eupolyphaga Steleophaga and compared with the medicinal materials(Y0,H0)before storage.The results are as follows:After 3 months of storage,Eupolyphaga Steleophaga with 10%water content have mildewed under moderate temperature,medium humidity,high temperature and high humidity,and the aflatoxin content exceeds the pharmacopoeial standard.The content of extract and cellolytic activity of the Eupolyphaga Steleophaga with3%water content under low temperature and low humidity were significantly higher than the other two groups(P<0.05),and the water content and crude fat content were significantly lower than the other two groups(P<0.05).Aspergillus flavus The total amount of toxins and the content of aflatoxin B1were also lower than the other two groups.In a low temperature and low humidity environment,the content of 3%water content and cellolytic activity of Eupolyphaga Steleophaga were significantly higher than that of 10%water content Eupolyphaga Steleophaga,and the total crude fat and aflatoxin were significantly lower than that of 10%water content Eupolyphaga Steleophaga(P<0.05).In summary,it is a better storage method for the Eupolyphaga Steleophaga to dry to a water content of 3%at4°C and 60%RH.Its medicinal extract content is high,the cellolytic activity is strong,and the crude fat and aflatoxin content are low.(5)Use the best feeding method and drying temperature screened out in(1)and(2)for the to-be-harvested E.sinensis Walker so that the crude fat content is 10.8%(G)and 9.6%(D),respectively,and placed at low temperature and low humidity(4℃,60%RH),medium temperature and humidity(25℃,65%RH),high temperature and high humidity(40℃,80%RH)environment,serial number is(G4、D4)、(G25、D25)and(G40、D40).After 3 months of storage,the samples were taken out to determine the internal quality and aflatoxin content of the medicinal materials of the Eupolyphaga Steleophaga and compared with the medicinal materials before storage(G0,D0).The results showed that after 3 months of storage,the extract content,anticoagulant and fibrinolytic activities of group D were higher than those of group G under the same storage conditions,and the total amount of aflatoxin was lower than that of group G under the same storage conditions.Among them,D4extract content and cellolytic activity were significantly higher than D25and D40(P<0.05),and water content,total aflatoxin content and aflatoxin B1content were all lower than D25and D40.In summary,the low-fat Eupolyphaga Steleophaga is a better storage method at 4℃and 60%RH.Its medicinal extract content is high,fibrinolytic activity is strong,and aflatoxin is low.
