| Background Bacterial vaginosis(BV)is a mixed infection caused by normal flora imbalance in the vagina and is the most common lower genital tract disease in women of childbearing age worldwide.Konjac glucomannan(KGM)is a water-soluble heteropolysaccharide isolated from konjac plant root powder.Studies have shown that KGM has prebiotic activity,anti-infection,anti-tumor and immunomodulatory functions.Specific Free Pathogen(SPF)Kunming female mice were used to study the effect of KGM on vaginal environment and gastrointestinal environment,and to provide experimental basis for KGM as a prebiotic and its further development and application in the treatment of bacterial vaginitis and restoration of vaginal microecosystem.Objective To investigate the prebiotic activity of KGM,combined with Lactobacillus reuteri to verify its effect on bacterial vaginosis.Methods Twenty-five female Kunming mice were randomly divided into 5 groups:normal group,model group,KGM group,Lr group,and konjac combined with Lactobacillus reuteri group.Five mice in each group,except the normal group,the other four groups of mice were treated with estradiol benzoate and suspension injection of phenol mucilage and mixed bacteria(E.coli:S.aureus:anti-ampicillin E.coli=5:4:1)to establish BV models,and throughout the experiment,200μL of 10~9CFU/m L konjac suspension and 50μL of vaginal administration were intragastrically administered daily in the konjac KGM group,200μL of10~9CFU/m L probiotic suspension and 50μL of vaginal administration were intragastrically administered daily in the L.reuteri Lr group,and 200μL of 10~9CFU/m L konjac probiotic suspension and 50μL of vaginal administration were intragastrically administered daily in the konjac combined with L.reuteri group.On day 15,the mice were anesthetized and sacrificed,and the relevant parameters were dissected and measured.Results(1)Compared with the normal group,the mice in the model group had decreased body weight,significant vaginal congestion and edema,increased number of E.coli and S.aureus,and decreased number of Lactobacillus.The expression of b Lacto m RNA in vaginal lavage fluid was significantly decreased,the expression of b Eco m RNA was significantly increased(P<0.05),the expression levels of TNF-αand IL-6 in vaginal uterine tissue were significantly increased(P<0.05),and the vaginitis model of surface mice was successful.(2)Compared with the model group,the vulvar clinical index scores of mice in the KGM,Lr,and konjac combined with probiotics groups were reduced to varying degrees,with the konjac combined with probiotics group decreasing most significantly(P<0.05).The number of E.coli and S.aureus and b Eco m RNA expression in vaginal lavage fluid showed a decreasing trend(P<0.05),and the number of Lactobacillus as well as b Lacto m RNA showed an increasing trend(P<0.05)in the KGM,Lr,and konjac combined probiotic groups.The expression levels of TNF-αand IL-6 in the KGM,Lr,and konjac combined probiotic groups were decreased to varying degrees(P<0.05).Conclusion It is concluded that konjac glucomannan combined with Lactobacillus reuteri has anti-inflammatory therapeutic effect on bacterial vaginosis in mice,improves vaginal environment and promotes vaginal health,and the mechanism of action may be related to its prebiotic effect and anti-inflammatory effect. |
