Objective: By observing the sciatic nerve of neuropathic pain due to chronic compression model rats,compared to the building and the temperature before and after acupuncture treatment before and after the general situation and behavior change,and to study abnormal expression of m RNA in rat hypothalamus,biological functions and path analysis of its target genes.To study the effect of Mongolian medical warm acupuncture mechanism,provide new theoretical basis for Mongolian medical warm acupuncture treatment.Methods: 32 SD rats were randomly divided into: Sham operation group,model group,warm acupuncture group and western medicine group,8 rats in each group.After 1 week of adaptive feeding,the model was made on the 8th day by Bennett and Xie method.The sham operation group only exposed the ischium nerve without ligation,and the other steps were the same as the model group.The treatment began on the 15 th day after modeling,and the sham operation group and the model group were not treated,and only normal saline was injected into the abdominal cavity.The warm acupuncture group was treated with Heyi point and Shen point,warm acupuncture therapy on a daily basis and intraperitoneal injection of raw brine.The western medicine group was given amitriptyline(0.1 g/kg)intraperitoneal injection.Made the day before die.The general behavior of rats was observed one day before modeling and 7,14,21,28 days after modeling,and the mechanical stimulation threshold(MWT)and thermal contraction leg threshold(TWL)were measured.After the treatment,collected in the rat hypothalamus organization,from the control group,model group,the warm acupuncture group randomly selected from four rats with high throughput screening of differentially expressed m RNA sequencing technology,target genes projections for significant differences in expression of m RNA,GO on target genes function enrichment and KEGG pathway analysis.Excavated with sciatica and cognitive impairment are closed signal pathway and using QPCR technology to detect targets related target genes,so as to verify the high-throughput sequencing results.Results: 1.General conditions: There was no significant difference in body weight,water intake,feeding amount,feces,hair,behavior and action of mice in each group before modeling(P>0.05).After modeling,the hair color of the rats decreased,the activity decreased,and they limped when walking,occasionally the limbs could not be lifted to the ground.With the prolongation of modeling time,the above symptoms were aggravated,and the rats often licked the claws,threw the claws and even self-mutilation.After treatment,compared with the model group,the above symptoms in the sham operation group,the warm acupuncture group and the western medicine group were significantly alleviated.2.The mechanical stimulation threshold(MWT)and thermal contraction threshold(TWL)in the behavioral experiment had no significant difference among the groups before modeling(P>0.05),but after modeling,the thresholds in the model group,the warm acupuncture group and the western medicine group were significantly lower than those in the sham operation group(P<0.05).Compared with the model group,the threshold value of the sham operation group,the Mongolian medical warm needling group and the western medicine group increased significantly after treatment(P<0.05).3.Transcriptome sequencing results: There were 361 differentially expressed m RNAs in sham-operated group compared with model group,of which 227 m RNAs were up-regulated and 134 m RNAs were down-regulated.There were 215 differentially expressed m RNAs in the model group compared with the warm acupuncture group,of which 57 were up-regulated and 158 were down-regulated.GO enrichment was mainly concentrated in axon development and membrane potential after protrusion.The enrichment of KEGG pathway mainly concentrated in the neural active ligand-receptor interaction pathway,GABA signaling pathway,WNT signaling pathway and so on.4.QPCR results: Compared with WAM group,the expression of SLC6A12 in model group was significantly decreased(P<0.01),and the expression of ADCY1 and GABRD was significantly increased(P<0.01),which proved to be consistent with the results of transcriptome sequencing.Conclusion: Mongolian medical warm acupuncture therapy can promote the circulation of heyi and blood,relieve pain and dredge the white pulse of CCI rats,thus improving the symptoms of dyskinesia,emotional instability and cognitive impairment.The specific mechanism of action may be influenced by multiple biological functions such as axonal development of target genes and the involvement of pathways such as GABA and WNT,thereby affecting the development of chronic neuropathy. |
