Objective : The Mongolian warm acupuncture therapy was used as the treatment method.The morphological changes of the posterior cervical muscle and the protein expression of Bax,Bcl-2,Caspase-3,Caspase-9 and the effect of apoptosis were observed by transmission electron microscopy,TUNEL assay and immunohistochemistry.The effectiveness of Mongolian warm acupuncture therapy on chronic injury model rats of cervical muscle was verified,and its mechanism was explored from a biological perspective.Provide scientific basis and new ideas for clinical syndrome differentiation.Methods : 36 healthy male Wistar rats were selected,and 10 were randomly selected as the blank group.The blank group did not do any treatment as a control.In addition to the blank group,all rats were modeled by chronic injury of cervical muscles.After confirming the success of the model,the rats were divided into groups for intervention treatment.Warm acupuncture group : acupuncture ’ Zhenhui point ’,1 time / day,15 min / time,western medicine group : meloxicam tablets were given by gavage;model group : only fixed without treatment.After the end of the intervention treatment,the neck muscle tissue of each group of rats was taken,and part of it was used for transmission electron microscopy to observe the morphological changes of rat neck muscle.In the other part,the apoptosis rate of rat posterior cervical muscle cells was detected by TUNEL method and the protein expression of Bax,Bcl-2,Caspase-3 and Caspase-9 in rat posterior cervical muscle cells was detected by immunohistochemistry.The experimental data were analyzed by SPSS 26.0 software to analyze the experimental data.One-way analysis of variance was used to describe all the data in the form of mean ± standard(X ±S).P < 0.05 indicated that the difference between the groups was statistically significant.Results : 1.Results of transmission electron microscopy : In the blank group,the myofibrils of the posterior cervical muscle were arranged regularly and closely,the structure was clear,the Z-line structure was normal,and there was no fracture.The sarcoplasmic reticulum structure was clear and no widening was observed.In the model group,the myofilaments of the posterior cervical muscle were disordered,the gap of myofibrils was widened,some Z lines were broken and disordered.In the warm acupuncture group,the myofibrils of the posterior cervical muscles of the rats were closely arranged,the structure was clear,the Z-line structure was normal,no fracture was observed,the sarcoplasmic reticulum structure was clear,and no widening was observed.Meloxicam western medicine group : the myofibrillar gap of the posterior cervical muscle of rats was widened,and some Z lines were broken.2.TUNEL test results :Compared with the blank group,the apoptosis of the model group was significantly increased(P < 0.01).Compared with the model group,the number of apoptotic cells in the warm acupuncture group and the western medicine group decreased,with statistical significance(P < 0.05).3.Immunohistochemical test results : Compared with the model group,the expression of Bcl-2in the cervical posterior muscle cells of the blank group was more than that of the model group(P < 0.01),and the expression of Bcl-2 in the cervical posterior muscle cells of the warm acupuncture group and the western medicine group was relatively increased(P < 0.05).Compared with the model group,the expression of Bax,Caspase-3 and Caspase-9 in the posterior cervical muscle cells of the blank group was relatively small,which was statistically significant(P < 0.01).In the comparison between the warm acupuncture group,the western medicine group and the model group,the expression of Bax,Caspase-3 and Caspase-9 in the cervical posterior muscle cells of the warm acupuncture group and the western medicine group decreased,with statistical significance(P < 0.05).Conclusion : 1.The Mongolian warm acupuncture therapy can effectively inhibit the apoptosis of the posterior cervical muscle tissue in the model rats with chronic cervical muscle injury and improve the tissue morphology of the posterior cervical muscle after chronic cervical muscle injury2.Mongolian warm acupuncture therapy can effectively reduce the apoptosis rate of cervical muscle tissue in rats with chronic cervical muscle injury.The mechanism may be related to the promotion of Bcl-2 expression and the inhibition of Bax,Caspase-3 and Caspase-9 protein expression.
