ObjectiveUHRF1(ubiquitin-like with PHD and ring finger domains 1,including the ubiquitin-like protein of PHD and RING finger 1 domains)is a nuclear protein gene related to cell growth.Its function is mainly involved in DNA repair and tumor Occurrence and development.This experiment mainly detects the expression of UHRF1 protein in benign and malignant pleural effusions,and explores the diagnostic efficacy of UHRF1 on malignant pleural effusions.MethodsIn the course of this study,37 cases of benign pleural effusion and 54 cases of lung cancer with malignant pleural effusion were selected as the research objects.Among them,the former included 5 cases of chronic obstructive pulmonary disease,12 cases of inflammatory disease,3 cases of empyema,and 17 cases of tuberculosis.The latter includes 3 cases of small cell lung cancer,10 cases of squamous cell carcinoma and 41 cases of adenocarcinoma.The specimens were collected from the First Affiliated Hospital of Gannan Medical College from March 2020 to January 2021.Both groups of specimens were centrifuged to extract the supernatant in a uniform method,and the expression of UHRF1 in each group was detected by enzyme-linked immunosorbent assay.And analyze whether there is a difference in their expression in the two groups of pleural effusions,and finally analyze whether it is related to the patient’s gender,age,smoking history,tumor location,histological type,etc.SPSS was used to process the data,and the obtained UHRF1 expression in each group was expressed by the mean ± standard deviation.The independent sample t-test was used to analyze the patient’s age,gender,smoking history,and tumor location on UHRF1 in malignant pleural effusions.Whether the expression is affected or not,one-way analysis of variance(ANOVA)was used to analyze the expression of UHRF1 in adenocarcinoma,squamous cell carcinoma,and small cell lung cancer in histological classification.The Youden index is used to determine the best cut-off value of UHRF1,the receiver operating characteristic curve(receiver operating characteristic,ROC)is used,and the area under the ROC curve(AUC)is used to explore the effect of UHRF1 protein expression on malignant pleural effusions.The diagnostic efficiency of the solution is compared with carcinoembryonic antigen(CEA),neuron-specific enolase(NSE),cytokeratin-19 fragment(Cyfra21-1),and the specificity and sensitivity of the four are tested.ResultsIn the malignant pleural effusion group,the expression of UHRF1 protein was significantly higher than that of the benign pleural effusion group.According to the results of comparative analysis,the difference between the two groups was very obvious.By studying the ROC curve,we know that the area under the curve corresponding to UHRF1,CEA,NSE,and cyfra21-1 are 0.791,0.830,0.759,and 0.615,respectively,and the sensitivities are 53.70%,66.70%,87.00%,72.20%,and the specificities are respectively.They are 91.90%,75.70%,59.50%,51.40%.Analysis of UHRF1 and clinical pathological parameters of patients showed that the expression level of UHRF1 protein was not different in different gender,age,tumor location,smoking history and histological classification(P>0.05).ConclusionThe results of this experiment show that the UHRF1 gene is significantly different between the benign and malignant pleural effusion groups,which means that in the clinical diagnosis of benign and malignant pleural effusion,UHRF1 can be used as a reference basis to jointly detect CEA in pleural effusion The expression of NSE,Cyfra21-1 can improve the diagnostic efficiency of clinical diagnosis of lung cancer and reduce the misdiagnosis rate.UHRF1 is expected to become a new tumor marker for the diagnosis of malignant pleural effusion.
