| Objective:According to the results of Global Cancer Prevalence Statistics(GLOBOCAN),breast cancer has become the malignant tumor with the highest morbidity and mortality rate in women,accounting for about 1/4 of all malignant tumors.In this study,we investigated and analyzed the mechanisms and clinicopathological features related to the occurrence of breast cancer in this region based on the biochemical analysis method and functional validation experiments,aiming to provide new objective research data for early diagnosis,individualized targeted therapy and prognosis assessment of breast cancer,and contribute to the prevention and treatment of breast cancer in Dali region.Methods:1.To define the study sample using statistical analysis to determine the high prevalence of pathological tissue types of breast cancer in the Dali region;2.Differentially expressed genes(DEGs)were screened using analysis based on GEO database,GEO2R online tool and Venn mapping tool,and the DEGs were screened with the use of STRING and Cytoscape to identify key genes that were more correlated with breast cancer,and then and the key genes were analyzed by GO analysis and KEGG pathway analysis through DAVID website,and the expression difference between breast cancer tissues and paraneoplastic tissues was analyzed by GEPIA,with P<0.05 as statistically significant;then the key genes were analyzed by Kaplan-Meier Plotter for prognostic survival,with P<0.05 as statistically significant,and the finally resultant genes as core genes;3.1-2 core genes were selected,and IHC and Western Blot experiments were performed to verify the protein expression of the selected core genes in breast cancer tissues and normal breast tissues,then analyze their relationship with clinicopathological characteristics and survival prognosis to generate relevant research results.Results:1.It was concluded that the pathological tissue type of high-incidence breast cancer in the Dali region was invasive ductal carcinoma,accounting for 84.86%,which was taken as the study subject for the follow-up study;2.Based on bioinformatics analysis,a total of 13 key genes were screened,which were PLIN1,PCK1,ADIPOQ,PDK4,CD36,FABP4,GPAM,CFD,LIPE,LEP,CIDEC,LPL,and PPARG,and after using differential expression analysis and survival prognosis analysis,five core and prognosis-related genes were finally identified as PLIN1,ADIPOQ,CFD,LPL,and PPARG;3.CFD and LPL genes were selected to functional validation experiments were performed using IHC and Western Blot experimental techniques,and the differences between their expression and clinicopathological features were analyzed using SPSS25.0software,and the following findings were observed:(1)IHC results showed that the expression of CFD and LPL in invasive ductal carcinoma of the breast were both significantly lower than normal breast tissue(P<0.05),and the study suggested that the development of breast cancer was associated with low expression of CFD and LPL,and the same result was suggested by the results of Western Blot experiments of CFD;(2)Low expression of CFD was significantly different in patients with invasive ductal carcinoma of the breast in terms of age(<51 years)and BMI(<24 kg/m~2)and presence of a family history of tumor(P<0.05),suggesting that the development of breast cancer is associated with metabolic disturbances due to immune factors;(3)There was a significant difference between the low expression of LPL and the increase of Ki-67 index(P<0.05);(4)CFD showed a significant positive correlation with the low expression of LPL in breast invasive ductal carcinoma(r=0.291,P<0.05),revealing a correlation between the development of breast invasive ductal carcinoma.Conclusions:1.The high prevalence pathological type of breast cancer in Dali region was invasive ductal carcinoma;2.The expression of CFD and LPL in breast invasive ductal carcinoma were both lower than that in normal breast tissue and were statistically significant(P<0.05);3.Low expression of CFD was statistically associated with age,BMI index,and family history(P<0.05);4.Low expression of LPL was statistically associated with a higher Ki-67 index(P<0.05);5.Positive correlation between CFD and LPL expression in breast invasive ductal carcinoma tissue(r=0.291,P<0.05). |
