| Objective: 1.To observe the efficacy and safety of Yizhi honey ointment(YZ)in the treatment of patients with amnestic mild cognitive impairment.2.The effects of YZ on memory,anti-fatigue and anti-hypoxia in healthy and carrier model mice were observed.Methods: 1.Clinical observation of YZ on patients with mild cognitive impairment The amnestic mild cognitive impairment(a MCI)patients in the internal medicine clinic and inpatient of Kashgar Uygur medical hospital were screened.72 patients with astringent mucinous a MCI were randomly assigned to compound muniziqi granule + YZ(experimental group)and donepezil hydrochloride(control group)according to the random number table.Observe the main and secondary symptoms of Uygur medicine syndrome;Adas cog score,MMSE score and logical story memory scale score were performed at the time of admission,12 th week and 24 th week;All cases were examined by ECG,blood routine,urine routine and stool routine.2.Animal experimental study on YZ improving memory function:(1)Effect on memory and endurance of healthy mice.50 male SPF KM mice were randomly divided into blank group,positive control group,YZ low dose,medium dose and high dose groups.The effects of YZ on memory,anti-fatigue and anti-hypoxia in healthy mice were observed by intragastric administration,platform jumping test,weight-bearing swimming test and hypoxia tolerance test.The changes of liver glycogen and muscle glycogen after administration and the content of whole blood lactic acid after exercise were measured.(2)Effects on learning and memory ability of scopolamine induced memory impaired mice: 60 male SPF KM mice were randomly divided into 6 groups: blank group,model group,positive drug(donepezil hydrochloride)group,YZ low dose,medium dose and high dose groups,with 10 rats in each group.They were administered by gavage for 15 days.From the 9th day of administration,1 hour after administration,the model was made by intraperitoneal injection of 2 mg / kg scopolamine,and the blank group was injected with equal volume of healthy saline.After administration,maze experiment and platform jumping experiment were carried out,and the pathological changes of brain hippocampus and cortex were observed;The contents of acetylcholine(ACh),acetylcholinesterase(ACh E),reduced glutathione(GSH),malondialdehyde(MDA)and superoxide dismutase(SOD)were measured in hippocampus.Results: 1.Clinical observation of YZ on patients with mild cognitive impairment :Comparing the curative effects of two groups of Uygur medicine syndrome,YZ can effectively improve Uygur medicine syndrome in patients with mild cognitive impairment,and there is a significant difference between main and secondary symptoms(P<0.05);The scores of MMSE,ADAS cog and lmiib in the experimental group and the control group at 12 and 24 weeks after treatment were higher than those at the time of admission.There were significant differences before and after treatment(P<0.05);The results of repeated measurement analysis of variance showed that the scores of MMSE,ADAS cog and limbi in the experimental group and the control group were not statistically significant(P > 0.05).2.Animal experimental study on YZ improving memory function:(1)Effects on memory and endurance of healthy mice: 1)YZ could significantly prolong the latency of platform jumping mice(P<0.001)and reduce the number of platform jumping errors(P<0.001);2)Compared with the blank control group,the swimming time of YZ high dose group was significantly prolonged(P<0.01);3)Compared with the blank control group,the hypoxia resistant survival time of mice in YZ high dose group was significantly prolonged(P<0.001);4)The measurement of hepatic glycogen showed that the content of liver glycogen in YZ groups increased significantly(P<0.001).The measurement of muscle glycogen showed that compared with the blank control,the muscle glycogen reserve of YZ groups increased significantly(P<0.05);5)The whole blood lactic acid content of mice in low,medium and high dose YZ groups was significantly lower than that in the blank control group immediately and 60 minutes after weight-bearing swimming(P<0.001);(2)Effects on learning and memory of memory impaired mice: 1)YZ medium and high dose groups could reduce the percentage of residence time in the new forearm and the number of entering the new forearm of scopolamine induced memory impairment mice(all P<0.05);2)YZ could reduce the number of errors in platform jumping experiment of memory impaired mice caused by scopolamine(P<0.01);3)YZ medium and high dose groups could inhibit ache enzyme activity in mouse brain tissue(P<0.05);YZ high dose group could increase the content of GSH in brain tissue of mild memory impairment model mice(P<0.05);YZ medium and high dose groups could increase the content of SOD and reduce the content of MDA in the brain of mild memory impairment model mice(all P<0.05);4)Pathological sections showed that the structures of hippocampus and cortex in the blank group were intact.In the model group,the number of cortical cells decreased,the focal cells in hippocampal C3 area were loose and the number of cells decreased,which was the pathological basis of behavioral disorder.The number of cortical cells and hippocampal cells decreased in YZ low-dose group,but the structure was slightly complete compared with the model group.The hippocampal and cortical structures of positive group,YZ medium dose group and YZ high dose group were intact.Conclusion: YZ can improve the Uygur medical symptoms and clinical symptoms of patients with mild cognitive impairment.No obvious adverse reactions were found during the treatment.Its curative effect is not inferior to that of donepezil hydrochloride control group.The results of animal experiments showed that YZ could improve the memory,anti-fatigue and hypoxia tolerance of healthy mice;In the animal model of memory impairment,YZ can significantly eliminate free radicals,improve the pathological structure of cerebral cortex and hippocampus,slow down the aging of nerve cells,and significantly improve the memory ability of animals with memory impairment. |
