| Objective: By detecting the expression of ciRS-7,miR-7 and HDAC8 in malignant melanoma tissues and surrounding paracancer tissues.Explore the relationship among the three,and to provide a new clinical scheme for the diagnosis and treatment of malignant melanoma by analyzing the correlation between these three factors and clinical and pathological parameters such as clinical staging,Breslow depth and surface ulcers.Methods: Firstly,the expression of ciRS-7,miR-7 and HDAC8 in malignant melanoma tissues and its surrounding paracancer tissues was detected by qRT-PCR method.In addition,the expression of HDAC8 protein in malignant melanoma tissues was detected by immunohistochemical method.The correlation between the expression of ciRS-7,miR-7 and HDAC8 was preliminarily investigated,at the same time,the relationship between these three factors and the clinical and pathological parameters of patients with malignant melanoma was analyzed.Finally,all data were verified by statistical methods(p≤0.05 was considered statistically significant).Results:(1)The results of qRT-PCR experiment showed that: ciRS-7,miR-7 and HDAC8 were expressed in malignant melanoma tissues and adjacent paracancer tissues,but the m RNA expression of ciRS-7 and HDAC8 was up-regulated in malignant melanoma tissues,and the expression of miR-7 was decreased,which was statistically significant compared with that in paired paracancer tissues(P < 0.05).(2)miR-7,HDAC8 and ciRS-7 were associated with clinical staging,Breslow depth and surface ulcers of malignant melanoma(P<0.05)were not correlated with sex,age,lymph node metastasis,distant metastasis,or clark grade(P>0.05),the difference was statistically significant.(3).ciRS-7 was negatively correlated with miR-7 expression in malignant melanoma tissues(r =-0.984,P < 0.0001),ciRS-7 was positively correlated with the expression of HDAC8(r=0.973,P<0.0001).(4).Immunohistochemical results indicated that HDAC8 protein could be expressed positively or strongly in both the nucleus and cytoplasm of malignant melanoma tissue,with a positive and strong positive rate of87.5%.In paraccarcinoma tissue,HDAC8 protein could be expressed in the nucleus,with a positive and strong positive rate of 52.5%,showing significant difference(P<0.05).Conclusion:In malignant melanoma,ciRS-7 is negatively correlated with the expression of miR-7 and positively correlated with the expression of HDAC8.Therefore,based on the regulatory axis of ciRS-7/miR-7/HDAC8,The up-regulated expression of HDAC8,the target gene of miR-7,may be caused by the competitive binding of ciRS-7,which may be involved in the occurrence of malignant melanoma,and the overexpressed HDAC8 may be transferred from the nucleus to the cytoplasm in some way,thus playing a regulatory role in the occurrence and development of malignant melanoma.The combined detection of these three factors has auxiliary significance for the staging diagnosis and targeted treatment of malignant melanoma in clinical work. |
