Objective:1.To explore the expression and significance of miRNA-21 regulating PDCD4 in malignant melanoma tissues.2.Predicting and authenticating the target gene of miRAN-21,researching the impact of miRNA-21 on PDCD4 expression.3.Researching the impact of miRNA-21 on proliferation and apoptosis of malignant melanoma.Method:From January 2016 to August 2016,20 cases of malignant melanoma and 20 cases of healthy skin tissues removed by skin surgery in Qilu Hospital of Shandong University were selected,and the expressions of miRNA-21 and PDCD4 m RNA in malignant melanoma and healthy skin tissues were detected by real-time PCR.Western blot was used to detect the expression of PDCD4 protein in malignant melanoma tissues and healthy skin tissues.Human malignant melanoma A375 cells were cultivated in cultivation box,so that passage was digested with digestive juice.The technology of luciferase reporter gene was used to prove that PDCD4 was the downstream target gene of miR-21,A375 cell was transfected,and total protein was extracted for Western blot experiment to detect the expression change of PDCD4 protein.CCK-8 method was used to detect the multiplication capacity of A375 cells.Hoechst 33258 staining method was used to detect apoptosis of A375 cells.real-time PCR was used to detect the expression of PDCD4 m RNA in A375 cells,and western blot method was used to detect the expression of PDCD4 protein in A375 cells.Result:Compared with the healthy control group,the expression level of microrna-21 in lignin melanoma is regulated by sup(P<0.05).The expression level of PDCD4 m RNA was down regulated(P<0.05).The expression of PDCD4 protein was significantly downregulated(P<0.05).PDCD4 is the target gene of miR-21.After miR-21 expression was inhibited,the cell reproductive capacity of A375 apparently decreased,and the apoptosis rate of A375 cells obviously increased.It could be seen that miR-21 had an inhibition action on the expression of PDCD4.Conclusion:The levels of miRNA-21 and PDCD4 m RNA in malignant melanoma tissues were significantly increased,while the levels of PDCD4 m RNA and protein were significantly decreased,which may be involved in the pathogenesis of malignant melanoma.miR-21 can participate in the course of attack of malignant melanoma through regulating the expre-ssion of the downstream target gene PDCD4. |
