| Objective: To observe the hypoglycemic and lipid-lowering effects of ethanol extract of Cichorum glandulosum Boiss.et Huet.(CG)in different doses on type 2 diabetes db/db mice and the effects on intestinal flora.Methods: 40 db/db mice were randomly divided into 4 groups,with the same number of mice in each group,which were respectively named as model group(MOD group),metformin treatment group(MET group),low dosee ethanol extract of CG(CGL group),and high dose ethanol extract of CG(CGH group).Another 10 m/m mice were selected as normal control group(CON group).In each treatment group,alcohol extract of CG and metformin solution were prepared with distilled water every day for intragastric administration.The CGL and CGH were100mg/kg/d and 200mg/kg/d respectively.Metformin was used as a positive drug to participate in the efficacy evaluation,and the dose was 200mg/kg/d,which was continuously gavaged for 8weeks.The daily intake of water was recorded,and the weekly fasting blood glucoseand body weight were measured.After treatment,oral glucose tolerance test(OGTT)and insulin tolerance test(ip ITT)were carried out.The mouse feces were collected into sterile centrifuge tubes,part of which was used for 16 Sr RNA sequencing ofintestinal flora,and the rest was determined by GC method.After the treatment of mice,the blood was taken from the eyeball and the serum was separated for the detection of glucose and lipid metabolism and other related indicators,and the liver,white fat of epididymis and brown fat of scapula were taken out for weighing,and the visceral index was recorded.Some of them were fixed with 4% paraformaldehyde formorphological observation,and the rest were frozen for the detection of theexpression of brown key protein of white fat by the Western Blot test.Results: After 8 weeks of CG administration,the hyperglycemia and hyperlipidemia of db/db mice were improved to some extent.The experiment observed that CG administration could reduce the blood glucose and lipid levels of db/db mice;enhance the glucose and insulin tolerance of db/db mice,increase insulin sensitivity;significantly improve the insulin resistance of mice.CG enhanced the T-AOC,SOD,GSH,CAT of the liver tissue of db/db mice;reduced the size of lipid droplets and the shape of white fat cells in the liver cells of db/db mice;up-regulated the key proteins of white fat browning UCP-1 and PGC-1 expression;increase the content of SCFAs in the feces of db/db mice,increase the intestinal hormone GLP-1 and inprove the body insulin resistance.The intestinal microflora sequencing showed that after administration of CGH,the intestinal microflora structure of db/db mice could be changed,reducing Bacteroide,Prevotellaceae,Lactobacillaceae,Bacteroidaceae,Alloprevotella,Prevotella,Bifidobacterium,Bacteroides acidifaciens.The results of correlation analysis showed that there was a significant negative correlation between lactobacillacea and TG,TC;Bacteroidaceae was negatively correlated with TG and HOMA-IR;There was a significant negative correlation between Firmicutes and LDL.Conclusion: CG has the effect of reducing blood glucose and blood lipids in db/db mice,change the abundance of intestinal flora in db/db mice;It can promote the browning process of white fat in db/db mice and improve the body insulin resistance,which can be used for the prevention and treatment of T2DM. |
