| Objective:By treating alveolar echinococcosis cultured in vitro and alveolar echinococcus cultured in C57BL/6mouse peritoneal cavity with Sunitinib and albendazole,we hope that the combination of Sunitinib and albendazole can further improve the inactivation ability of alveolar echinococcus.Methods:1.Alveolar Echinococcus protoscoleces were extracted and their activity was identified using eosin staining.In vitro experiments were divided into 5 groups: Blank group(blank control group),DMSO group(solvent control group),25 μg /m L ABZ group(Albendazole powder group),50 ng /m L SU11248 group(Sunitinib powder group),12.5 μg /m L ABZ+25 ng /m L SU11248 group(Albendazole powder combined with Sunitinib powder group).Eosin staining was used to identify and calculate the survival rate of echinococcosis.2.The alveolar echinococcosis in C57BL/6 mice were treated with drugs.After 60 days of culture,they were randomly divided into 5 groups: Blank group(Control group),Control group(infection control group),ABZ group(Albendazole powder group),SU11248 group(Sunitinib powder group),ABZ+SU11248 group(Albendazole powder combined Sunitinib powder group),with 8 rats in each group.C57BL/6 mice were intraperitoneally modeled with extracted alveolar echinococcosis.60 days after modeling,the mice in each group were given 100 mg/kg albendazole and 50 mg/kg Sunitinib by intragastric administration,once a day.In combination group,one drug was given in a single day,ABZ and SU11248 were given alternately,and no treatment was given in Control group.Mice in each group were euthanized after 60 days of treatment,and tissues such as alveolar echinococcosis,liver and blood were taken from the abdomen.3.The alveolar echinococcosis tissues in the abdominal cavity of C57BL/6 mice in each group were weighed,and the cyst inhibition rate was calculated.The serum levels of alanine aminotransferase(ALT/GPT)and aspartate aminotransferase(AST/GOT)were detected by ALT/GPT and AST kits.The levels of IL-2 and IL-4 in serum of mice were detected by ELISA kits.The expression of CD4 and CD34 in the alveolar echinococcosis tissues of the liver and the peritoneal cavity were detected by immunohistochemistry.Results:1.ABZ+SU11248 group showed better in vitro inhibitory effect on Echinococcosis alveolaris than single drug administration mode.2.The peritoneal hydatid infection model of mice was established successfully.The wet weight of hydatid cyst in ABZ+SU1124 group was lower than that in ABZ group,SU11248 group and Control group,and the cyst inhibition rate in ABZ+SU11248 group was higher than that in ABZ group(P < 0.05).3.Compared with the Blank group,groups of mice serum ALT,AST level have different degree rise,the Control group were significantly increased;Compared with the Control group,each treatment group mice serum ALT,AST expression level is significantly lower;The expression levels of ALT and AST in ABZ and SU11248 combined group were lower than those in ABZ and SU11248 groups(P < 0.05).4.ELISA showed that serum levels of IL-2 and IL-4 in ABZ+SU11248 group increased significantly after treatment compared with other groups.5.Immunohistochemical experiments in mice alveolar spines ball larva tissues for detecting level of CD34 and CD4: vesicular spines ball larva tissues in mice abdominal cavity infection ABZ CD4 +SU11248 group level for high expression,higher than ABZ,SU11248 group and Control group(P < 0.05).CD4 level in SU11248 group was higher than that in ABZ group,ABZ+SU11248 group and Blank group(P < 0.05).CD34 expression level of ABZ+SU11248 group was lower than that of ABZ group and Control group in liver alveolar hydatid tissue and abdominal hydatid tissue(P < 0.05).Conclusion:Sunitinib can inhibit the growth of alveolar echinococcosis by inhibiting angiogenesis.The combination of Sunitinib and albendazole has a stronger inhibitory effect on the activity of alveolar echinococcosis and can reduce the liver damage caused by alveolar echinococcosis than the two treatments alone. |
