| Objective: to study the effects of acupuncture on lipids and Homocysteine of Hyperlipidemic Rats with Phlegm-Dampness stagnation type basing on Aryl hydrocarbon receptor-CD36 pathwayMethods: 45 male wistar rats were randomly divided into 3 groups,there are control group,model group and acupuncture group,respectively.Rats of model group were feed with high-fat diet,rats of acupuncture group were treated by acupuncture at pishu point,xuehai point,sanyinjiao point and zusanli point.Sera of rats were collected and then detected by ELISA assay,including lipids indexes,anti-oxidant indexes,Homocysteine(Hcy)and S-adenosyl methionine(SAM).Protein and RNA of liver tissue were extracted and then detected by western blot and RT-q PCR,respectively.Including CD36 and aryl hydrocarbon receptor(AHR).Oil red O staining was used to visualize the deposition of lipids in liver.Results: compared with model group,total cholesterol(TC)(P<0.01),total triglyceride(TG)(P<0.05),high density lipoprotein(HDL)(P<0.05)and Hcy(P<0.05)were decreased in acupuncture group,while low density lipoprotein(LDL)and oxidized low density lipoprotein(ox-LDL)were increased(P<0.05).Compared with model group,sera hepaticlipase(HL)and SAM in acupuncture group were elevated(P<0.05),while MDA(P<0.05)and FAS(P<0.01)were decreased.The results of western blot and RT-q PCR showed that compared with control group,the expression level of CD36 and AHR were up-regulated in model group,while compared with model group,the expression level of CD36 and AHR were down-regulated(P<0.05).according to oil red o staining,acupuncture treatment could prevent the deposition of lipids in liver of rats.Conclusion: Acupuncture at pishu point,xuehai point,sanyinjiao point and zusanli point could decrease the level of lipids and Hcy in sera of rats,moreover,acupuncture treatment also has an anti-oxidant effect and prevent lipids from depositing in liver,the underlying mechanism might be achieved by AHR-CD36 pathway. |
