Objective: Using "TCM Inheritance Auxiliary Platform(V2.5)",systematically explore the core drugs of chronic heart failure and Yang deficiency,summarize the rules of the drugs,and provide data basis for new drug research and development.Methods: The CNKI,Wan Fang,VIP and Pubmed database system from January1980 to December 2021,were screened according to the inclusion and exclusion criteria,and summarized their therapeutic drugs.The "TCM Inheritance Auxiliary Platform(V2.5)" was used to analyze the relevant data of drugs and summarize their drug use rules.Using network pharmacology technology,the TCMSP and ETCM databases were searched to screen the components of core drugs and obtain their targets.The Pubchem database was used to supplement potential targets,and Uniprot standardized target names.Use Gene cards,Drug bank,TTD database to capture disease targets,and then use Venny platform to capture drug and disease target network and protein interaction network through STRING database and Cytoscape 3.6.0 software,use MCODE plug-in to select core targets;use GO and KEGG analysis to display the results on We Chat platform.Using Pubchem,PDB database download the first four core components and target structure,the Open Babel software into mol2 format,using the open source version of Py Mol software to select the core target protein and component structure to water molecular operation,using Auto Dock software for receptor and ligands,the target protein as a receptor,components as a ligand,semi-flexible way.Molecular docking data were visualized using an open-source version of the Py Mol program.Results:1.A total of 357 effective prescriptions were obtained,involving 194 traditional Chinese medicines.The top 10 frequencies were aconite,poria cocos,Salvia miltiorrhiza,Astragalus,Angelica,cassia,diarrhea,licorice and white peony root.The Four gases are mainly warm;the five flavors are mainly sweet.The return meridian is mainly attributed to the spleen meridian.8 Core drugs: aconite,Poria cocos,Salvia miltiorrhiza,astragalus,Atractylodes,cassia branch,lung chestnut,diarrhea disease.10 pairs of high frequency drug combinations with the highest frequency and 14 pairs of core drug combinations with the highest confidence and 6potential new prescriptions.2.Through network pharmacology,a total of 257 active components,283 drug targets,2,036 disease targets,and 157 drugs and disease intersection targets were identified from 8 core drugs.A total of 23 core target proteins were screened out.The results of GO analysis show that it is related to the regulation of gene expression,the inhibition of cell apoptosis and other processes.The results of KEGG analysis,mainly involving the PI3K-Akt signaling pathway,the Lipid and atherosclerosis pathway,etc.3.Molecular docking results,the docking between core components and target sites is generally good,especially the docking performance between quercetin and each other.Conclusion: 1.The basic principle of clinical treatment of chronic heart failure(Yang deficiency syndrome)is to invigorate qi and warm Yang,promote blood circulation and improve water.2.TCM treatment of Yang deficiency type of chronic heart failure may play a role by controlling cardiomyocyte apoptosis,promoting neovascular regeneration,and affecting lipid metabolism.3.Molecular docking showed that the drug core components and can better combination between core targets,including quercetin may play a major role in the process of treatment.
