Objectives:Polycystic ovary syndrome(PCOS)is a common endocrine disorder and metabolic syndrome in women.The study aimed to evaluate the relationship between kisspeptin levels and metabolism-related clinical indicators,and to conduct a metabolomics study on serum studies of PCOS patients with different kisspeptin levels and non-PCOS patients to explore the relationship between serum kisspeptin levels and PCOS metabolic abnormalities.Methods:Clinical studies and metabolomics studies:1.Collect clinical data and blood samples of 143 patients with PCOS and 131 patients with non-PCOS,and detect serum kisspeptin and metabolism-related indicators by ELISA;2.PCOS patients were grouped according to kisspeptin concentration,and serum metabolites were analyzed by using class-targeted metabolomics of high kisspeptin PCOS(H),low kisspeptin PCOS(L)and non-PCOS(C).Results:1.BMI,WHR,blood pressure,hypertrichosis and acne scores in PCOS were significantly higher than non-PCOS group;in PCOS group,kisspeptin,LH,FSH,LH/ FSH,T,FPG,TG,TC,and LDL-C levels were significantly higher than non-PCOS group;PCOS patients with different kisspeptin levels were significantly different in2 h blood glucose,FNS,HOMA-IR,AUCG,AUCI,HDL-C,FFA,BA,and LEP levels.2.Correlation analysis showed that serum Kisspeptin levels of 274 study subjects were positively correlated with general clinical indicators BMI,WC,WHR,m F-G score,acne score,and sex hormone markers LH,FSH,LH / FSH,and T(P <0.05),with FPG and TG,and HDL-C(P <0.05);in PCOS patients(n=143),kisspeptin was positively correlated with FNS,2h blood glucose,HOMA-IR,and AUCG.3.The serum metabolomic analysis showed that,Compared with the L group,Six metabolites were significantly upregulated in group H,87 metabolites were significantly downregulated,These differential metabolites are mainly involved in glycerol oil metabolism,amino sugar and nucleotide sugar metabolism,galactose metabolism,etc.Compared to the group of C,And 88 metabolites were significantly up-regulated in group H,The 53 metabolites were significantly downregulated,These differential metabolites are mainly involved in arginine biosynthesis,pantothenate and Co A biosynthesis,fatty acid degradation,etc.Of the 131 metabolites were significantly upregulated in group L compared to group C,17 metabolites were significantly downregulated,These differential metabolites are mainly involved in propionate metabolism,pyruvate metabolism,fructose and mannose metabolism,arginine biosynthesis,valine,leucine and isoleucine degradation,citric acid cycle,aminoglyand nucleotide sugar metabolism,etc.Conclusions:1.Kisspeptin correlation analysis further verified the involvement of kisspeptin in the occurrence of PCOS.2.Kisspeptin may be involved in the abnormal glucose and lipid metabolism.3.Kisspeptin may be involved in the regulation of serum metabolites of PCOS,suggesting that kisspeptin may be associated with the metabolic abnormalities of PCOS.
