OBJECTIVE:To explore the value of image analysis of CD10 immunophenotype for the clinicopathological diagnosis and differential diagnosis of hepatocellular carcinoma.METHODS:Hepatocellular carcinoma cases were collected from the Department of Pathology of Sanya Central Hospital and the Department of Pathology of the First Affiliated Hospital of Hainan Medical College.Group A consisted of 29 cases of well-differentiated hepatocellular carcinoma with corresponding paraneoplastic liver tissues;Group B consisted of 127 cases of hepatocellular carcinoma tissues(including 98 cases of well-differentiated hepatocellular carcinoma and 29 cases of medium-low differentiated hepatocellular carcinoma).The wax blocks of all cases were made into immunohistochemical sections and then made into digital images,the immunophenotypic characteristics of CD10 in the images of both groups were collected and recorded using image analysis software.The collected measurement index data were examined and progeny were included in the one-way screening with binary logistic regression analysis to derive the percentage of correct prediction and binary logistic equation for each group,and the measurement indexes with significant correlation with diagnostic results were screened.RESULTS:Group A: There were eight CD10 immunophenotypes with significant correlation with the diagnostic results,the most important measure was the mean optical density of the CD10 staining area.The values of the measured indicators were substituted into the binary logistic equation and 96.6% correct diagnostic compliance rate was obtained.Group B: There were eight CD10 immunophenotype measures that were significantly correlated with the diagnostic results,the most important measure was the number of CD10 staining areas.The values of the measured indicators were substituted into the binary logistic equation,and 92.1% correct diagnostic compliance rate was obtained.CONCLUSION:The combination of image analysis and immunohistochemistry can broaden the perspective of pathologists and obtain morphological features that have certain reference significance for clinical pathology work,so that pathologists can more accurately and comprehensively understand the morphological changes of diseases.The most important measurements of immunohistochemical CD10 in distinguishing well differentiated hepatocellular carcinoma from paraneoplastic liver tissue and hepatocellular carcinoma with different degrees of differentiation are: average optical density of stained areas,and number of stained areas,respectively.
