Research background and purpose:Lung cancer is one of the malignant tumors that seriously affect human health all over the world.with the development of China’s chemical industry,increasingly serious environmental pollution and the increase in national demand for tobacco,the incidence of lung cancer has been rising in recent years.its mortality has leapt to the top of all malignant tumors.Non-small cell lung cancer(non-small cell lung cancer,NSCLC)is the most common type of lung cancer.The main histological types are adenocarcinoma and squamous cell carcinoma.The prognosis of lung cancer is highly related to the early detection and treatment of the disease.Surgical resection is still recognized as the most effective method at present.With the rapid development of medical technology,remarkable progress has been made in the diagnosis and treatment of lung cancer,but due to the lack of obvious symptoms and signs of early lung cancer,there are still many patients with regional or distant metastasis at the time of diagnosis.in the late stage of the disease,lost the opportunity of operation,the clinical prognosis is poor.Related studies have shown that the 5-year survival rate of NSCLC is only 15%.Therefore,to explore the related molecular mechanism of lung cancer and new molecular therapeutic targets,and to improve the early diagnosis rate and prognosis of patients with lung cancer are still very important for the future diagnosis and treatment of lung cancer.In this study,paraffin specimens of NSCLC were selected as the object of study.The expression of PRDX2(peroxredoxin2,PRDX2)protein and RUNX2(runt-related transcription factor 2,RUNX2)protein in NSCLC was detected by immunohistochemical method,and the relationship between the expression of these two proteins and the clinicopathological features and prognosis of NSCLC was analyzed,and the correlation between PRDX2 and RUNX2 protein in NSCLC was discussed.Materials and Methods:A total of 78 NSCLC specimens(45 adenocarcinomas,33 squamous cell carcinomas,47 males and 31 females)were selected from the Department of Cardiothoracic surgery of the first affiliated Hospital of Chengdu Medical College from January 2018 to December 2018.NSCLC cancer tissues were used as the experimental group and the corresponding paracancerous tissues as the control group.Immunohistochemical EnVision method was used to detect the expression of PRDX2 and RUNX2 in 78 cases of NSCLC carcinoma and corresponding adjacent normal tissues;The expression of PRDX2 and RUNX2 proteins in NSCLC and their relationship with clinicopathological features and prognosis of NSCLC were analyzed by SPSS25.0 statistical software,and the correlation between the expression of these two proteins was discussed.Result:1.In 78 cases of NSCLC carcinoma and corresponding adjacent normal tissues,the high expression rate of PRDX2 protein was 61.54%in NSCLC carcinoma tissues and 24.36%in corresponding adjacent normal tissues,and the high expression rate was 61.54%and 24.36%in corresponding normal tissues,respectively.RUNX2 protein was highly expressed in 52 cases of NSCLC carcinoma,with a high expression rate of 66.67%(52±78),and in 23 cases of adjacent normal tissues,the high expression rate was 29.49%(23±78).The expression levels of PRDX2 and RUNX2 in NSCLC carcinoma tissues were significantly higher than those in adjacent normal tissues(P<0.05).2.Among the 78 cases of NSCLC,there were significant differences in the expression of PRDX2 among different smoking history groups,tumor pathological types,lymph node metastasis groups and TNM staging groups of NSCLC(P<0.05):The high expression rate of tissue protein in smoking patients was higher than that in non-smoking patients,and the protein expression rate in patients with squamous cell carcinoma was higher than that in patients with adenocarcinoma.The protein expression rate in patients with lymph node metastasis was higher than that in patients without lymph node metastasis,and the protein expression rate in patients with stage Ⅱ-Ⅲ was higher than that in patients with stage Ⅰ.There was no significant difference in PRDX2 among different age,sex and tumor differentiation groups(P>0.05):The expression of PRDX2 was not related to age,sex and tumor differentiation.3.In 78 cases of NSCLC,the expression of RUNX2 was significantly different among different pathological types of NSCLC,lymph node metastasis and TNM stages(P<0.05):The expression rate of protein in patients with adenocarcinoma was higher than that in patients with squamous cell carcinoma,and the expression rate in patients with lymph node metastasis was higher than that in patients without lymph node metastasis.the protein expression rate in patients with stage Ⅱ-Ⅲ was higher than that in patients with stage Ⅰ.However,there was no significant difference in RUNX2 among different age,sex,smoking history and tumor differentiation(P>0.05):The expression of PRDX2 was not related to age,sex,smoking and tumor differentiation.4.There was a positive correlation between the expression of PRDX2 and RUNX2 protein in NSCLC(r=0.391,P<0.05).5.Patients with high expression of PRDX2 and RUNX2 proteins had significantly shorter survival time and worse prognosis(P<0.05).Conclusions:1.The expression of PRDX2 protein and RUNX2 protein in non-small cell lung cancer was significantly higher than that in paracancerous tissues,and both of them may be involved in the occurrence and development of non-small cell lung cancer.2.The increased expression of PRDX2 protein may be related to smoking history,lymph node metastasis,TNM stage and pathological classification of lung cancer;The increased expression of RUNX2 protein may be related to lymph node metastasis,TNM stage and pathological classification of lung cancer.3.There is a positive correlation between the expression of PRDX2 and RUNX2 in lung cancer.They play a synergistic role in the occurrence and development of non-small cell lung cancer and may participate in the proliferation and distant metastasis of non-small cell lung cancer.4.The expression of PRDX2 and RUNX2 in lung cancer may have prognostic value in patients with non-small cell lung cancer.
