| Objective: To investigate the expression of Supervillin and IL-1β in epithelial ovarian cancer(EOC)by mining the differential proteins in EOC using bioinformatics tools,to analyze the clinicopathological relationship between them and EOC,and to explore the correlation between them in the process of EOC occurrence and development.Methods: Using EOC data from the Clinical Proteomic Tumor Analysis Consortium,GO and KEGG analysis are used to understand the biological functions and inflammation-related processes of EOC differential proteins;survival curves and ROC curves are plotted using R language and prognostic analysis is performed to screen for differential proteins.Immunohistochemistry is used to detect Supervillin and IL-1β in40 cases of EOC group(22 cases of ovarian epithelial serous carcinoma,6 cases of ovarian epithelial mucinous carcinoma,6 cases of endometrioid carcinoma and 6 cases of clear cell carcinoma),20 cases of epithelium-derived ovarian borderline tumor group(9 serous borderline tumor and 11 mucinous borderline adenoma)and 20 cases of ovarian benign control group(12 ovarian mucinous cystadenoma,8 ovarian serous cystadenoma)are expressed.Results: The results of GO analysis shows that EOC differential proteins are mostly enriched in the biological process of cell-substrate adhesion;in cellular localization,differential proteins are more present in focal adhesion and cell-substrate junction;in molecular science,differential proteins arre mainly involved in actin binding;the results of KEGG enrichment analysis suggests that differential proteins could be enriched in the process of regulation of actin of cytoskeleton.Goplot results indicats that EOC differential proteins are involved in regulation of cell junction assembly,including Supervillin;further screening of differential proteins combined with clinical data shows that there is a difference in overall survival between the high and low expression groups of Supervillin protein in EOC(p < 0.05);the area under the ROC curve of Supervillin protein is greater than 0.7.Multivariate independent prognostic analysis suggests that age,histological grade and Supervillin high expression could be independent prognostic factors(p < 0.05),with histological grade and Supervillin expression as high risk factors(HR > 1).Immunohistochemical results suggest that the positive expression rates of Supervillin and IL-1β in EOC are higher than those in benign controls,and the differences are statistically significant(P < 0.05);the positive expression rates of Supervillin and IL-1β in EOC tissues are correlated with the degree of tissue differentiation and clinical stage,and the differences are statistically significant(P < 0.05),and with age,histological type,presence or absence of ascites and lymph node metastasis are not significantly correlated(p > 0.05);their expression in EOC is positively correlated(r = 0.373,p = 0.018).Conclusion: Supervillin and IL-1β may be involved in ovarian carcinogenesis,and they have synergistic effects.In addition,Supervillin can better predict the survival and prognosis of ovarian cancer,and may become a biological marker and therapeutic target for ovarian cancer prognosis. |
