Objective: To investigate the relationship between iron metabolism and oxidative stress in periodontitis through bioinformatics analysis and clinical experiments.Methods:1.The differentially expressed genes related to iron metabolism and periodontitis were screened from GEO、OMIM and Genecards databases,and the GO function and KEGG enrichment analysis of the differentially expressed genes were performed using DAVID database,the differentially expressed genes were mapped into PPI protein network interactions using STRING online and cytoscape 3.9.1 analysis software.2.According to the diagnostic criteria for periodontitis,a total of 34 patients were enrolled in the department of stomatology of Xinjiang Uygur Autonomous Region People’s Hospital,including 15 patients with periodontitis and 19 patients with periodontal health,and collecting clinical periodontal indexes、gingival crevicular fluid(GCF)and venous blood.The periodontal indexes such as PD and BI、the concentration of MDA in GCF and the content of SI、SF and TRF in serum were detected,to compare the difference between periodontitis group and Healthy group.Results: 1.Differentially expressed genes are involved in the development of periodontitis through inflammatory and immune responses.2.Compared with the healthy group,PD and BI in periodontitis group were significantly increased(P<0.001).3.The number of residual teeth in periodontitis group was lower than that in healthy group(P<0.05).4.The serum SI and SF contents in periodontitis group were significantly higher than that in healthy group(P<0.05).5.There was no significant difference in TRF between periodontitis Group and healthy group(P>0.05).6.The content of MDA in GCF of periodontitis group was higher than that in healthy group(P<0.05).Conclusion:1.There is an interaction in differentially expressed genes between periodontitis and iron metabolism,and PD,BI are positively correlated with SI and SF,indicating that the degree of periodontal inflammation is correlated with abnormal iron metabolism,which might be related to the pathogenesis of periodontitis.2.Compared with healthy group,MDA content in GCF of periodontitis group increased,suggesting that oxidative stress tended to increase.
