It is the dried persistent calyx or persistent calyx with fruit of the perennial herb Physalis alkekengi L.of the family Solanaceae,which is cold in nature,bitter in taste and belongs to the lung meridian.It is also known as Luo Shen Zhu,Hanging Golden Lantern and Red Mushroom Maiden Fruit.Modern pharmacological studies have shown that the plant has significant pharmacological activities such as antibacterial,anti-inflammatory,anti-tumour,antioxidant,anti-diabetic and diuretic.The alkaloids of Physalis alkekengi L.have been rarely studied,therefore,this thesis provides a reference for the further development and utilization of the alkaloids.The specific research content and experimental results are as follows:In this paper,the alkaloids of Physalis alkekengi L.were firstly determined by systematic pre-experimentation of the active ingredients in the mallow.The results showed that the extraction rate of total alkaloids obtained by the ultrasonicassisted extraction method was much higher than that of the other extraction methods;based on the single-factor experiments,the optimal extraction process number of alkaloids was determined as follows: material-liquid ratio 1:20 g/m L,ultrasonic time 32 min,ethanol volume fraction 91%,reflux extraction time 1.6 h.The extraction rate of total alkaloids reached 0.169 g/kg.The purification process and parameters were further tested to compare the static adsorption and desorption capacities of three different types of macroporous adsorbent resins,D-101,NKA-9 and AB-8,using the adsorption and desorption rates as indicators,and the results showed that the NKA-9 macroporous resin had the best adsorption and desorption performance for the total alkaloids of the brome.The optimum p H value for the alkaloids was 9.0,the mass concentration was 4.2466 mg/m L and the sample volume was7 BV,followed by 14 BV of ethanol at 75% by volume.The alkaloids were concentrated under reduced pressure after resin purification and then repeatedly recrystallized with isopropanol at an optimum dosage of 30 m L.After recrystallization,the alkaloids were stored at 0-4°C for 12-15 h and crystals were obtained by static crystallization.The structure of the alkaloids of Physalis alkekengi L.was analysed by UV spectroscopy,IR spectroscopy,high performance liquid chromatography.The characteristic absorption peak of the mallow alkaloids was 416 nm by UV spectroscopy,and it could be identified as alkaloids by combining the absorption of the characteristic functional groups by IR spectroscopy.The High-performance liquid chromatography analysis confirmed that one of the alkaloids was atropine sulfate.Finally,the antibacterial effect and in vitro antioxidant activity of the alkaloids of Physalis alkekengi L.were investigated.The inhibition effect of the alkaloids on Escherichia coli,Staphylococcus aureus and Streptococcus thermophilus was shown to be significant,with the lowest inhibition concentrations of 5,20 and 80 mg/m L for each species respectively.The alkaloids of Physalis alkekengi L.has strong reducing ability,and can effectively scavenge DPPH and ABTS free radicals,and has strong antioxidant ability.
