Effects Of Jiaji Electroacupuncture And Sports Running Wheel On Motor Function And SOD,GSH,Glu Levels In Rats With Spinal Cord Injuries

Objective:The models of spinal cord transection injuries were established in this research.This study mainly observed effects of jiaji electroacupuncture and sports running wheel on motor function and SOD,GSH,Glu levels in rats with spinal cord injries.Probed into the mechanism of action about treatment of jiaji electroacupuncture and sports running wheel in the spinal cord injuries.Method:Ten-week-old adult female SPF rats were selected.The total number was 72.According to the random number table method,these rats were divided into sham-operated group,model group,j iaj i electroacupuncture group,jiaji electroacupuncture and sports running wheel group.All the rats were fed for a week adaptively.Then the rats in sham-operated group were took away laminas,but the spinal cords were not excised.The rats’spinal cords of T10 were excised in model group,j iaj i electroacupuncture group,j iaj i electroacupuncture and sports running wheel group.After the models were made smoothly,all of the rats were adopted to ususl care.On the third day,the rats in jiaji electroacupuncture group,jiaji electroacupuncture and sports running wheel group were treated accordingly.All of the rats were observed their BBB locomotion scores as behavioral scores on day 7,14,28.All of the rats were detected the levels of SOD,GSH by ELISA in the areas of spinal cord injuries.All of the rats were detected the expression of Glu by LC-MS/MS in the areas of spinal cord injuries.Results:Behavioral assessment results:The rats’BBB locomotion scores were 21 in sham-operated group on day 7,14,28.The rats’BBB locomotion scores in model group were lower than that of sham-operated group on day 7,14,28(P<0.05).At the same time point,the rats’BBB locomotion scores were higher in jiaji electroacupuncture group,jiaji electroacupuncture and sports running wheel group than that of model group(P<0.05).At the same time point,The rats’BBB locomotion scores were higher in jiaji electroacupuncture and sports running wheel group than that of jiaji electroacupuncture group(P<0.05).Compared within groups,The rats’BBB locomotion scores were higher in jiaji electroacupuncture and sports running wheel group at different time point(P<0.05).The results of the levels of SOD,GSH by ELISA in the areas of spinal cord injuries:The expression of SOD,GSH in sham-operated group didn’t show obvious change(P>0.05).Compared to the sham-operated group,the expression of SOD,GSH in model group was lower than the sham-operated group at the same time point(P<0.05).Compared to the model group,the expression of SOD,GSH in jiaji electroacupuncture group,j iaj i electroacupuncture and sports running wheel group were higher than model group at the same time point(P<0.05).Compared to the jiaji electroacupuncture group,the expression of SOD,GSH in jiaji electroacupuncture and sports running wheel group were higher than jiaji electroacupuncture group at the same time point(P<0.05).Compared within groups,the expression of SOD,GSH in jiaji electroacupuncture and sports running wheel group was increased markedly at different time point(P<0.05).The results of the concentration of Glu by LC-MS/MS in the areas of spinal cord injuries:The concentration of Glu in sham-operated group didn’t show obvious change(P>0.05).Compared to the sham-operated group,the level of Glu in model group was increased obviously at the same time point(P<0.05).Compared to the model group,the concentration of Glu in jiaji electroacupuncture group,jiaji electroacupuncture and sports running wheel group were decreased markly at the same time point(P<0.05).Compared to the jiaji electroacupuncture group,the concentration of Glu in jiaji electroacupuncture and sports running wheel group was decreased obviously at the same time point(P<0.05).Compared within groups,the concentration of Glu in jiaji electroacupuncture and sports running wheel group was decreased markly at different time point(P<0.05).Conclusion:1.Jiaji electroacupuncture,Jiaji electroacupuncture and sports running wheel can increase BBB scores and improve motor function in rats with spinal cord injuries.The therapeutical effect of Jiaji electroacupuncture and sports running wheel was better than that of Jiaji electroacupuncture.2.Jiaji electroacupuncture and sports running wheel can improve motor function visibly in rats with spinal cord injuries.The mechanism may be ralated to increase levels of SOD,GSH to relieve oxidative stress,and decrease levels of Glu to reduce excitatory amino acid toxicity.
