Data Mining And Network Pharmacology Study Of Traditional Chinese Medicine In The Treatment Of Chronic Atrophic Gastritis With Intestinal Metaplasia

Objective:Modern data mining technology was used to make statistics on the prescriptions of traditional Chinese medicine for chronic atrophic gastritis accompanied by intestinal metaplasia;explore the core prescriptions for the treatment of chronic atrophic gastritis accompanied by intestinal metaplasia;analyze their medication rules,and then discuss the possible etiology and pathogenesis of the disease to guide clinical medication.On this basis,the network pharmacology research method was used to analyze the possible mechanism,in order to provide reference for the basic research and clinical application of the disease.Methods:Study 1:The clinical literature of traditional Chinese medicine in the treatment of chronic atrophic gastritis with intestinal metaplasia published in the relevant databases was searched.After screening according to the inclusion criteria of the literature,the literature was sorted and standardized into Excel table.The frequency of herb,four qi,five flavors,meridian tropism and efficacy of the herb were statistically analyzed.Association rule analysis and systematic cluster analysis of high-frequency traditional Chinese medicine were carried out,and the core herb combinations were screened out,and the clinical medication rules were analyzed.Study 2:The core herb components and targets were searched on TCMSP,TCMID and other platforms,and the intersections with the targets of chronic atrophic gastritis with intestinal metaplasia were imported into Cytoscape 3.8.0 to construct the network of traditional Chinese medicine-active ingredients-targets,and the topological values were analyzed to obtain the core compounds.The drug-disease intersection targets were imported into the STRING database to establish the PPI network,and the core targets were obtained.The data were input into Metascape,and GO and KEGG pathway enrichment analysis were performed to predict the target and related important signaling pathways of traditional Chinese medicine combination in the treatment of chronic atrophic gastritis with intestinal metaplasia.Results:A total of 267 articles were included in this study,and 267 prescriptions were recorded,including 211 Chinese herbs.Data mining analysis:(1)Sex,taste,and Channel tropism:The frequency of warm herbs was the highest,followed by cold herbs.The five flavors of the medicine were bitter,sweet and spicy.It mainly belongs to the spleen meridian,lung meridian,stomach meridian and liver meridian.(2)Herbs’ efficacy and frequency:The main efficacy of the herb is tonifying deficiency,followed by promoting blood circulation and removing stasis.Gancao,Baizhu,Danshen,Fuling,Banxia,Danshen,Baihuasheshecao,Baishao,Huangqi,and Ezhu were the top 10 drugs with the frequency from high to low.And the first 9 herbs were roughly consistent with the results of core herbs obtained by association rule analysis network diagram,which was the core prescription.(3)Association rule analysis:The minimum conditional support was set as 20,and the minimum rule confidence was 90%,and then get the core herb portfolio:B aizhu-Huangqi-Dangshen and B aizhu-Dangshen-Fulin.(4)Cluster analysis:Five clusters were obtained by systematic cluster analysis,including Banxia Xiexin Decoction,Chaihu Shugan Powder,Sijunzi Decoction,etc.Network pharmacological analysis:(1)Herb component and target:Core herb combinations obtained 63 active ingredients,and including 7 active ingredients of Baizhu,20 of Danshen,15 of Fuling and 21 of Huangqi;207 of herb targets.(2)Disease target:A total of 912 disease targets and 87 drug-disease intersection targets were obtained by database query.(3)Herb-active ingredients-target network and the topology analysis,and PPI:the core traditional Chinese medicine was Huangqi;quercetin,kaempferol and luteolin were the top three compounds;and IL6,MMP9 and TP53 were the top three core targets.(4)GO and KEGG enrichment analysis:the core herb combination treatment of chronic atrophic gastritis with intestinal metaplasia may have positive regulation of gene expression,negative regulation of apoptosis process,positive regulation of cell proliferation,response to lipopolysaccharide and other biological processes.Possibly Pathways in cancer,lipid and atherosclerosis,PI3K-Akt signaling pathway,prostate cancer,MAPK signaling pathway play a therapeutic role.Conclusion:1.The core prescriptions of modern Chinese medicine in the treatment of chronic atrophic gastritis with intestinal transformation are Gancao,Baizhu,Danshen,Fuling,Banxia,Danshen,Baihuasheshecao,Baishao,Huangqi.The main treatment idea is to intonate the spleen and stomach,inactivating blood circulation and removing blood stasis,and detoxizing dampness.The pathogenesis of the disease may be related to the weakness of spleen and stomach and the accumulation of blood stasis and dampness in the stomach.2.Traditional Chinese medicine may act on core targets such as IL6,MMP9 and TP53 through quercetin,kaempferol,luteolin and other components,and play anti-inflammatory,anti-tumor and protective effects on gastric mucosa,and its mechanism may be related to PI3K-Akt signaling pathway,and MAPK signaling pathway.
