| Objective:By detecting NOD-like receptor family,pyrin domain-containing protein 4(NLRC4)in plasma of patients with Chronic Heart Failure(CHF),and Interleukin-1β(IL-1β)to explore the relationship between inflammatory response and the occurrence and development of CHF.Method:1.A total of 165 patients with CHF(NYHAII ~ IV),II(n = 45),III(n = 63),IV(n = 57),and 35 healthy controls(CON)were selected.The concentrations of IL-1β and NLRC4 in plasma were detected by ELISA,and the protein expressions of NLRC4 and IL-1β were detected by Western Blot.2.General clinical data including age,sex,blood glucose,liver and kidney function were collected and compared.The biochemical markers such as n-terminal pro-brain natriuretic peptide(NT-pro BNP),left ventricular ejection fraction(LVEF),left ventricular end-diastolic diameter(LVEDD)and left ventricular end-diastolic volume(LVEDV)were recorded in the laboratory and echocardiographic room of our hospital.3.SPSS was used for statistical analysis of general data.Graphpad prism and PS software were used for graphing analysis.Results:1.The expression levels of NLRC4 and IL-1β in CHF group were significantly higher than those in CON group(P<0.01).2.Pearson correlation analysis showed that NLRC4 and IL-1β were positively correlated with NT-pro BNP,LVEDD,LVEDV in each CHF group(r=0.335、0.191、0.425、0.353、0.333、0.532,P<0.01),was negatively correlated with LVEF(r=-0.331,-0.367,P < 0.01),NLRC4 was positively correlated with IL-1β(r=0.395,P<0.01).3.The ROC curve area of NLRC4 and IL-1β were 0.817 and 0.858 respectively,and the difference was statistically significant(P<0.01).4.The expressions of NLRC4 and IL-1β in peripheral blood mononuclear cells(PBMC)were detected by Western blot.Spearman correlation analysis showed that the expression levels of NLRC4 and IL-1β in plasma of CHF patients were positively correlated with cardiac functional grading(P<0.01).Conclusion:1.With the development of CHF,the levels of NLRC4 and IL-1β in the plasma of CHF patients increased gradually.The inflammatory reaction induced by NLRC4 and IL-1β plays an important role in the development of CHF.2.According to ROC curve analysis,NLRC4 and IL-1β have certain predictive value for chronic heart failure.3.The increased levels of NLRC4 and IL-1β in plasma of CHF patients were positively correlated with the severity of CHF,suggesting that the NLRC4 inflammasome may be associated with the development of chronic heart failure. |
