Objective:The purpose of this study is to explore the medication rules of ancient Chinese doctors in the treatment of phlegm-dampness infertility by Chinese Medicine Inheritance Auxiliary Platform(V2.5)software,and to predict the potential targets and mechanisms of core traditional Chinese medicine in the treatment of phlegm-dampness infertility based on network pharmacology.Methods:1.We collected and sorted out the internal-use formulas which are used to treat phlegm-dampness infertility from the Dictionary of Traditional Chinese Medicine Prescriptions(2nd edition),and standardized the data and introduced them into the Chinese Medicine Inheritance Auxiliary Platform software.Carries on the frequency statistics and the composition rule analysis to the statistical prescription,excavates the core medicine,and summarizes the medication rules of ancient chinese doctors in the treatment of phlegm-dampness infertility.2.The related targets of infertility were searched from GeneCards,OMIM and DisGeNET.Components and targets of core traditional Chinese medicine were retrieved from Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP).The intersection targets of the infertility and medicine were obtained by using the Venny2.1.0 online program.STRING was used to predict the protein-protein interactions.Gene Ontology(GO)term and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment of the key targets were performed by Metascape.Results:1.A total of 51 prescriptions were included in this study,involving 136 flavors of traditional Chinese medicine.The drugs with high frequency of use included Poria cocos,Pinellia ternata,Tangerine peel,Ligusticum chuanxiong,Angelica sinensis,Atractylodes macrocephala Koidz,Ginseng,Glycyrrhiza,and Xiangfu(Cyperi Rhizoma),etc.The properties and flavors of the drugs were mainly warm and octyl;meridian tropism mainly targeted spleen,liver and stomach.Based on drug association rule analysis,the drug combinations with top support rank were Poria cocos-Tangerine peel,Pinellia ternataPoria cocos,Pinellia ternata-tangerine peel,etc.Through prescription cluster mining and drug association rule analysis results,the core drugs were Poria cocos,Pinellia ternata,Tangerine peel,Ligusticum chuanxiong,Angelica sinensis,Atractylodes macrocephala Koidz,Ginseng,Glycyrrhiza,and Cyperi Rhizoma.2.Through the study of network pharmacology,it is predicted that the core drugs may act on 111 targets,including AKT1,TP53,IL-6,TNF,VEGFA,ESR1 and so on.The key targets were involved in the major biological processes of response to hormone,cellular response to lipid,gland development,response to decreased oxygen levels,response to nutrient levels and so on.KEGG pathway analysis showed that AGE-RAGE signaling pathway,HIF-1 signaling pathway,estrogen signaling pathway and NF-κB signaling pathway might play a key role in the treatment of infertility by core medicines.Conclusion:1.The properties and flavors of the main drugs used by ancient doctors to treat phlegm-dampness infertility are pungent and warm,and their meridian tropism mainly targeted spleen,liver and stomach.The treatment is mainly to invigorate the spleen and remove dampness,regulate qi and activate blood circulation.The core drugs were Poria cocos,Pinellia ternata,Tangerine peel,Ligusticum chuanxiong,Angelica sinensis,Atractylodes macrocephala Koidz,Ginseng,Glycyrrhiza,and Cyperi Rhizoma.2.The core traditional Chinese medicines are used to treat phlegm-dampness infertility by interfering with AGE-RAGE signaling pathway,HIF-1 signaling pathway,estrogen signaling pathway and NF-κB signaling pathway,regulating AKT1,TP53,IL-6,TNF,VEGFA,ESR1 and so on.They play a role in the treatment of phlegm-dampness infertility by improving chronic inflammation and oxidative stress,regulating glucose and lipid metabolism,promoting the development of oocytes and follicles,and improving ovulation disorders.Regulate ovarian secretory function,improve endometrial receptivity.
