| Objective:This study was conducted to investigate the effect of Tianyang pill on the proliferation of HepG2 cells through Hippo pathway by proteomics and in vitro experiments,and then to reveal the potential mechanism of its treatment of HCC glucose metabolism,to provide a scientific and theoretical basis for the clinical application of Tianyang pill,and to provide new ideas for the combined treatment of HCC with Chinese and Western medicine.Methods:1.HepG2 cells were cultured in vitro.2.The effect of different concentrations of Tianyang pill lyophilized powder on the activity of HepG2 cells was detected using CCK-8 assay to screen the optimal drug concentration.3.The toxicity effect of Tianyang pill on HepG2 cells was investigated by CCK-8method.4.The effect of Tianyang pill on the invasion and migration of HepG2 cells was investigated by Transwell assay.5.Probe the apoptotic mechanism of Tianyang pill on HepG2 cells by Annexin v-FITC/PI flow cytometry.6.Screen target genes by combined proteomics and bioinformatics.7.Detect the effect of Tianyang pill on the expression level of hippo-related pathway proteins by Western blot.8.Use kits to study the effect of Tianyang pill group and Hippo groups on the glucose metabolism of HepG2 cells.Results:1.compared with the control group,Tianyang pill significantly reduced the viability,proliferation rate,invasion rate and migration rate of HepG2cells(p<0.05);the results of annexin v-fitc/pi Alpine assay showed that Tianyang pill promoted HepG2 cell apoptosis(p<0.05).2.proteomics,raw letter analysis and western blot assay The results showed that Tianyang pill had an activating effect on Hippo pathway in hepatocellular carcinoma cells.3.The results of glucose metabolism showed that Tianyang pill exerted antihepatocellular carcinoma effect through Hippo pathway-mediated regulation of glucose metabolism.Conclusion:Tianyang pill can inhibit HepG2 cell proliferation,migration and invasion and induce apoptosis,and promote hippo pathway regulation of hepatocellular carcinoma glucose metabolism. |
