| Objective:To observe the clinical commonly used traditional Chinese medicine compound tianyang bolus of late Diabetic nephropathy(Diabetic nephropathy,DN)model rats of pharmacodynamics index(fasting glucose,glycosylated hemoglobin,urea nitrogen,creatinine,urine protein)and renal pathological changes and the effect of the herbal medicine compound pharmacodynamics effect are discussed.Based on the theory of "square evidence",this paper verified the symptoms of Yin and Yang and Yin and Yang deficiency in late DN by means of the method of "testing the evidence".Methods:60 male SD rats,rats were randomly divided into blank group and module,blank group use common feed,feed the rest of the group are high in fat and sugar 8 weeks after intraperitoneal injection of joint chain of small dose of urea with cephalosporins(Streptozocin,STZ),measurement of fasting plasma glucose values greater than 16.7 tendency for mmo/L,identified as Diabetes(Diabetes Mellitus,DM)model.The rats were divided into model group,positive drug group,dizi pill group and tianyang pill group according to the fasting blood glucose value.After 4 weeks,24 hours of urine was collected using the metabolic cage,and the urine microalbumin in rats was measured between 30 and 300mg/24h,and the replication of the DN model was considered successful.At this time,the positive drug group,the dizi pill group and the tianyang group respectively gave the corresponding drugs to the oral gavage,and continued to administer the drug for 8 weeks.Free drinking water during the experiment,without insulin and other hypoglycemic agents.At the end of the experiment,24 hours of urine was collected in rats to measure the 24-hour urine protein of rats.The blood glucose of the rats was examined by enzyme-linkedimm imosorbent assay(ELISA),and the indexes of fasting blood glucose,hemoglobin,urine protein,creatinine and urea nitrogen were detected in each group.After the rats were executed,the kidney was fixed by 4%poly formaldehyde solution,and HE staining was used to observe the pathological changes of the kidney in each group.The rest of the kidney is stored in liquid nitrogen.Results:1.The influence of the general situation of DN rats:compared with the blank group,model group rats activity decreased,unresponsive,depression,weight decrease,thin and fur lackluster,outlining the symptom such as bones protrude.The overall state of the rats in the positive drug group and the Dizi pill group was better than that of the model group,and the rats in the Tianyang pill group were the best compared with other drug intervention group rats.2.Influence of rat kidney index in each group:compared with the blank group,the renal index of rats in the model group increased significantly.Compared with model group,tianyang pill group,positive medicine group,the son of pellet group of DN rat kidney index values are lower,but the day rat kidney Yang pill group index reduced obviously,with statistical significance.3.Effect of fasting blood glucose on rats in each group:compared with the blank group,the fasting blood glucose values of rats in the other groups increased significantly.Compared with the model group,the fasting blood glucose values of each drug treatment group were decreased,and the fasting blood glucose was the lowest in the tianyang pill group,which was statistically significant compared with the positive drug group and the dizi pill group.4.Effect on the content of glycosylated hemoglobin in the group of DN rats:compared with the blank group,the content of glycosylated hemoglobin in rats in the remaining groups was significantly increased,and the difference was statistically significant.Compared with the model group,there was no statistically significant difference between the drug treatment groups.5.Effect of fasting insulin levels in the rats of the group of DN rats:compared with the blank group,the fasting insulin levels of the rats in the remaining groups were significantly increased,and the difference was statistically significant.Compared with the model group,the fasting insulin levels in the positive drug group,the dizi pill group and the tianyang pill group were all decreased,which was statistically significant.6.Effects on the levels of creatinine and urea nitrogen in rats:compared with the blank group,the levels of creatinine and urea nitrogen in the other groups were significantly increased.Compared with the model group,the levels of creatinine and urea nitrogen were significantly reduced in the positive drug group and tianyang pill group.Compared with the dizi pill group,the levels of creatinine and urea nitrogen were both decreased in tianyang pill group and positive drug group,and the lowest level of creatinine and urea nitrogen in the tianyang pill group was statistically significant.7.Effect of urine protein content of rats in each group:compared with the blank group,the contents of urine protein in the other groups were significantly increased.Compared with the model group,the urinary protein content of each drug treatment group decreased.Compared with the dizi pill group,the content of urine protein in the rats of tianyang pill group and the positive drug group decreased significantly,and the urine protein content of the rats in the positive group was the lowest,which was statistically significant.8.For each group of rat renal histopathological changes:after HE staining and observed under visible model group increased kidney size,glomerular ball diameter increases,the broadening of glomerular mesangial area,mesangial matrix mild nodular hyperplasia,expansion of renal tubule.After drug treatment,the renal pathological changes of rats were significantly reduced compared with those in the model group.Among them,the glomeruli and renal tubules of the rats in tianyang pill were slightly enlarged compared with that of the blank group,while the glomerular volume of the rats in the positive drug group was not significantly different from that of the rats.Conclusion1.Late day Yang pill can improve DN model rat’s general status,reduce relevant pharmacodynamics index,and effectively reduce urinary protein excretion,lower kidney hypertrophy index,reducing the level of glomerular sclerosis repair renal function,has certain protective effect to the kidney.2.On the basis of the concept of "square measurement with prescription",we compare the Tian Yang pill and the ground pill,and hope to provide modern experimental research and research data reference for the late DN development to the syndrome of Yin Yang and the two deficiency. |
