| Purpose Lung cancer is one of the most common cancers in the world,and approximately more than 60% of lung cancer patients are adenocarcinoma of the lung,despite promising therapeutic prospects for the clinical application of gene targeting and immune checkpoint inhibitors for mutations.Limited by the heterogeneity of different lung adenocarcinoma patients,treatment targeting specific genes can only benefit some patients,and lung adenocarcinoma remains one of the most clinically significant malignant diseases in terms of morbidity and mortality.Microtubule proteinε and δ complex 2(TEDC2)is involved in cell cycle regulation in a variety of tumors,and investigating the potential mechanism of its role may provide a new idea for clinical treatment of lung adenocarcinoma.Methods(1)Pan-cancer analysis of TEDC2 expression in multiple tumors based on UCSCXENA database expression profile files;(2)Obtaining clinical information from TCGA database and constructing cox regression models for correlation analysis of clinical diagnosis and survival prediction of TEDC2 in each tumor;(3)Bioinformatics analysis of correlation between TEDC2 and poor prognosis of lung adenocarcinoma;(4)Online database and q PCR to validate the protein and transcriptome level changes of TEDC2 in lung adenocarcinoma;(5)Online database to obtain the gene set of TEDC2 co-expression,analyze the expression profile files of TCGA lung adenocarcinoma patients,and extract the differential genes between high and low TEDC2 expression groups;(6)Collated the overlapping genes and corresponding expression differences of the above gene set,and further perform log transformation;(7)Functional enrichment of the collated gene profile files;(8)String database analysis of TEDC2 protein interactions with the overlapping gene sets and screening of the key genes;(9)TIMER and IOBQR package analysis of the relevance of TEDC2 to the immune system in the lung adenocarcinoma microenvironment;(10)Blocking of TEDC2 using si RNA,to analyze the altered biological functions of lung adenocarcinoma cells with down-regulated TEDC2 expression.Results(1)TEDC2 expression was elevated in multiple tumors and correlated with the prognosis of multiple tumors;(2)TEDC2 was highly expressed in lung adenocarcinoma and could be verified experimentally;(3)TEDC2 was associated with poor prognosis in lung adenocarcinoma;(4)gene enrichment analysis indicated that TEDC2 was involved in signaling pathways such as cell cycle,mitosis and natural immunity;(5)downregulation of TEDC2 partially blocked cell entry into division and reduced resistance to cisplatin,as well as reduced tumor migration and invasion,in addition to upregulating the expression of MHC II class molecules and LAG3.Conclusions Overexpression of TEDC2 is associated with poor prognosis in lung adenocarcinoma patients,and the possible mechanism of action is involved in cell cycle regulation and downregulation of MHC class II molecules resulting in malignant proliferation and immune escape of lung adenocarcinoma cells. |
