| Research Background&Objective:Lung cancer is currently the malignant tumor with the highest morbidity and mortality in the world.In 2020,there will be about 815,000 new lung cancer cases in my country,and about 710,000 deaths.The latest cancer data show that lung cancer is still a major disease that seriously threatens the health of the people.Non-small cell lung cancer is the most common histological type of lung cancer,accounting for more than 80%of lung cancer cases,of which lung adenocarcinoma is the main subtype,and surgery is the preferred treatment.In clinical practice,patients with lung adenocarcinoma show huge differences in the degree of benefit from treatment,which is directly related to the survival and prognosis of patients.This study aims to analyze the level of immune cell infiltration,differentially expressed genes,tumor TCR and BCR characteristics and related TCR sets in patients with lung adenocarcinoma.And the correlation analysis of TCR characteristics and immune infiltration,to explore the relationship between these analysis characteristics and prognosis.Methods:26 patients with lung adenocarcinoma diagnosed by pathology were collected.Tumor tissue and clinical data were collected by operation.RNA sequencing and immunologic library sequencing were performed.The sequencing results were combined with patient clinical data for a personalized bioinformatics analysis.Results:1.Gene enrichment analysis of GSEA results showed that patients with progressive disease were enriched on the p53 and TGF-β pathways,and patients with partial remission were enriched on the B cell related pathways.Genes located in this pathway are associated with phenotypic differences between groups.2.In the analysis of immune infiltration,patients with partial remission have higher B cell infiltration levels and lower Treg.Patients with partial remission and stable disease have slightly higher oligoclonal BCR than patients with progressive disease,and lower diversity than patients with disease progression.Progressive patient.3.The analysis of IFN,GEP and other expression scores is higher in patients with partial remission,and patients with higher IFN-y expression scores and GEP scores have higher TCR oligoclonal ability.4.Patients with progressive disease have higher TCR diversity,while patients with partial remission have fewer disease-related clones.5.Patients with higher CD4/CD8 ratios and higher cholesterol pathway expression scores have shorter progression-free survival(PFS),while patients with higher NLR(neutrophil to lymphocyte ratio)have longer Progressive survival(PFS).Conclusions:1.The CD4/CD8 ratio and NLR level can evaluate the prognosis of cancer patients.The cholesterol pathway is related to the prognosis of lung adenocarcinoma,which provides a research basis for further exploring the role of cholesterol pathway in lung adenocarcinoma.2.Patients with higher levels of B-cell infiltration are more likely to benefit from anti-tumor therapy.Immune repertoire technology can analyze the immune function status of patients with lung adenocarcinoma,and has important guiding significance in evaluating the efficacy and prognosis of lung adenocarcinoma. |
