| Objective Detect the expression levels of key factors of mi RNA-16-5p,Let-7a-5p,and ACE2-Ang(l-7)-MAS axis in essential hypertension(EH)Phlegm dampness abun dant syndrome group,non-EH Phlegm dampness abundant syndrome group,and hea lthy control group,and analyze the differences in their expression levels,To Explore the molecular biological characteristics of mi RNA-16-5p,Let-7a-5p,ACE2,Ang(1-7),and Mas in the EH group with phlegm dampness accumulation,non phlegm dampness accumulation,and healthy control group,providing a certain molecular biological basis for the syndrome essence and pathogenesis of primary hypertension with phlegm dampness accumulation.Methods 78 hospitalized patients with essential hypertension who visited the First Affiliated Hospital of Anhui University of Traditional Chinese Medicine and the Second Affiliated Hospital of Anhui University of Traditional Chinese Medicine from March2022 to October 2022(30 cases in the EH Phlegm dampness abundant syndrome group,and 48 cases in the non-EH Phlegm dampness abundant syndrome group)and 26 cases in the healthy control group were included.After signing the informed consent form,4ml of fasting venous blood was collected using an anticoagulant tube,and plasma was centrifuged to obtain mi RNA.The relative expression levels of mi RNA-16-5p and Let-7a-5p were measured using stem loop RT-q PCR;Use Elisa reagent kit to detect the concentrations of ACE2,Ang(1-7),and Mas.Result1.The relative expression level of mi RNA-16-5p : The EH group with excessive phlegm dampness was higher than the EH non phlegm dampness group(P<0.05);The EH phlegm dampness accumulation type group was higher than the healthy control group(P<0.05)2.The relative expression level of Let-7a-5p:The EH group with excessive phlegm dampness was higher than the EH non phlegm dampness group(P<0.05);The EH phlegm dampness accumulation type group was higher than the healthy control group(P<0.05)3.The levels of ACE2,MAS,and Ang(1-7): All of their levels were the lowest in the EH group with excessive phlegm dampness(P<0.05);All of their levels were the highest in the healthy control group(P<0.05).Conclusion The expression levels of Let-7a-5p and mi R-16-5p in the plasma of EH patients with excessive phlegm and dampness were higher than those of the healthy control group and non phlegm and dampness accumulation EH group;ACE2,Ang(1-7),and Mas have the highest levels in the healthy control group,and the lowest levels in EH with excessive phlegm dampness;The levels of Let-7a-5p,mi R-16-5p,ACE2,Ang(1-7),and Mas in plasma can serve as one of the biological markers for the diagnosis of EH phlegm dampness stagnation syndrome. |
