Objective:To study miRNA related to prognosis of colorectal cancer by bioinformatics and verify the role of miR-548e-3p in carcinogenesis by experiments.Methods:(1)In this study,colorectal cancer-related data were downloaded from TCGA(The Cancer Genome Atlas),including micro RNA(miRNA)data of each sample of colorectal cancer and clinical information of each sample.(2)From December 2021 to December 2022,cancer tissues and distal normal tissue samples of 25 colorectal cancer patients in the Affiliated Hospital of Youjiang Medical University for Nationalities were collected.The expression of miR-548e-3p from adjacent tissues was detected by RT-q PCR.(3)The transfection efficiency of miRNA and the expression level of E-cadherin were detected.(4)The expression levels of E-cadherin and N-cadherin in the miR-548e-3p inhibitor group and the normal control group were detected by western blot assay,and the scratch migration test and cell invasion test were carried out.Results:(1)Five kinds of miRNAs related to prognosis were screened by bioinformatics:miR-181a-5p,miR-548e-3p,891a-5p,miR-3189-3p and miR-3917.The relationship between miR-548e-3p and cancer has not been studied.(2)The content of miR-548e-3p in colorectal cancer and distal intestine tissues was detected by clinical specimens of colorectal cancer and distal intestine tissues.It was found that the expression level of miR-548e-3p in colorectal cancer tissues was significantly higher than that in distal intestine tissues.(3)The expression level of E-cadherin in the inhibitor group was higher than that in the control group and the expression level of N-cadherin was lower than that in the control group after 48 hours of transfection.In the scratch test,the rate of cell migration in miR-548e-3p inhibitor group was significantly lower than that in normal control group.In Transwell invasion test,the number of cells in the inhibitor group was significantly lower than that in the control group.Conclusion:(1)miR-548e-3p is highly expressed in cancer and correlated with the prognosis of colorectal cancer.(2)miR-548e-3p is closely related to invasion and migration of colorectal cancer and E-cadherin and N-cadherin.
