| Delphinium kingianum(Rannculaceae)belongs to the Ranunculaceae family and is distributed in southern Tibet.It usually grows in the mountains at an altitude of 4600 meters,and blossoms in August.Through literature investigation,it is found that its chemical constituents and their bioactivities have not been studied before.In order to search for compounds with novel structure and biological activity,this thesis conducted a systematic study on the chemical constituents of Delphinium kingianum for the first time.By ethanol extraction,acid extraction,alkali precipitation and solvent extraction,total alkaloids were prepared.Compounds were isolated and purified by by silica gel column chromatography.Twenty-three compounds were identified by various spectral techniques.Among them,compound 2-1*is a new compound named kingiadine,which is vakognavine C20-diterpenoid alkaloid.The known compounds include seventeen lycoctonine-type C19-diterpenoid alkaloid:gyalanine A(2-2),gyalanine B(2-3),methyllycaconitine(2-4),lycaconitine(2-5),anthranoyllycoctonine(2-6),ajacine(2-7),delavaine A free acid(2-8),delavaine B free acid(2-9),delsemine A(2-10),delsemine B(2-11),delavaine A(2-12),delavaine B(2-13),trifoliolasine B(2-14),browniine(2-15),lycoctonine(2-16),delectinine(2-17),delphatine(2-18),one hetisine-type C20-diterpenoid alkaloid:tangutimine(2-19),one isoquinoline alkaloids named(+)-glaucine(2-20)and three benzoic acid compounds:2-(4-methoxy-3-methyl-4-oxobutanamido)benzoic acid(2-21),2-(4-methoxy-2-methyl-4-oxobutanamido)benzoic acid(2-22)and methyl 2-(4-amino-2-methyl-4-oxobutanamido)benzoate(2-23).All of the compounds were isolated from Delphinium kingianum for the first time.In the biological activity test,lycoctonine,a compound isolated from Delphinium kingianum,showed a certain cardiac activity.After structural modification,8 derivatives were obtained.Subsequently,the cardiotonic activity of these derivatives were investigated by in vitro frog heart perfusion experiment.Preliminary results showed compound 3-13 exhibited obvious cardiotonic activity,and the growth rate of tension reached 118.09%when the concentration was 2mg/m L. |
