Objective:Changes in peripheral inflammatory factors IL-4,IL-6,IL-10,and TNF-αin umbilical Cord Blood Mononuclear Cells treated with ischemic stroke rats were investigated to explore the possible mechanisms in development.Methods:In 70 healthy male SD rats with a mass of between 250 and300 g.rats ratsrandomly divided into 3 groups for formal purposes,namely,Blood Mononuclear Cells(cb MNCs)group;saline group;pseudosurgery group.60 rats were treated with Transient Middle Cerebral Artery Occlusion(t MCAO).We excluded the rats that failed and died after making the mold and selected the rats that could be collected for all the complete time periods to get the final group:13 in cb MNCs Group;11 in saline group;10 in pseudosurgery group.In cb MNCs group,we injected rats with 5*10~6cells of umbilical cord blood mononuclear cells through the caudal vein after 24 hours molding,while saline group and pseudosurgery group were injected with 0.5 ml normal saline.Behavioral assessment of neural function in laboratory animals was performed at1,3,5,7 and 14 days after the modeling.Brain size of cerebral infarction was measured by TTC staining after 14days.Peripheral blood was collected from caudal vein 30min before injection,2h after injection,48h,72h and7days after molding,and serum concentrations of IL-4,IL-6,IL-10 and TNF-αwere measured by ELISA.Results:(1)TTC staining:Measureof size showed a significant decrease in infarct volume in cb MNCs group compared to the salinegroup;(2)m NSS:Compared with sham operation,the 24 h score of normal saline group was 8 to 16 points,indicating that the modeling was successfu.cb MNCs significantly improved m NSS scores from day 3compared to saline group,with statistically significant differences(P<0.05),and kept the same trend at day 5,day 7,and day 14.(3)Concentrations of inflammatory factors in peripheral blood of rats:The serum levels of IL-4,IL-10,IL-6,and TNF-αwere statistically significant(P<0.05)in saline group compared to pseudosurgery group within 24hours after the moulding.The serum concentrations of pro-inflammatory factors IL-6 and TNF-αdecreased slowly after reaching the peak at 48 h.Anti-inflammatory cytokines IL-4 and IL-10 continued to increase.cb MNCs group were compared to saline group at various time points,and there was no statistically significant difference between the two groups within 30 minutes before injection(P>0.05),while the pro-inflammatory factors IL-6 and TNF-αdecreased for the rest of the time(P<0.05)and the anti-inflammatory factors IL-4 and IL-10 increased(P<0.05).(4)Correlation analysis of the degree of recovery of nerve function defect and changes of inflammatory factors:Compared with normal saline group,pro-inflammatory factors IL-6 and TNF-αdecreased in cb MNCs group,while anti-inflammatory factors IL-4 and IL-10 increased.Inflammation was regulated,resulting in increased neurological function recovery and decreased brain tissue volume damage.Conclusion:Umbilical Cord Blood Mononuclear Cells can regulate inflammation after acute ischemic stroke and improve neurological function and brain tissue damage in laboratory animals.
