Objective:In this study,we investigated the various expression of DHX9 in clinical tissues of gastric cancer,normal adjacent tissues,normal gastric mucous epithelial cells and gastric cancer cells,and analyzed the possible clinical significance.In addition,we explored the effects of DHX9 on the proliferation,invasion and apoptosis of gastric cancer in vitro,and speculated on the potential mechanism.Methods:In this study,we explored the date from the TCGA database and investigate the expression of DHX9 in gastric adenocarcinoma.then we collected the tumor tissues and related clinical pathological data of gastric cancer patients to analyze the relationship between the expression of DHX9 and clinical pathological characteristics by using SPSS statistical software.The Kaplan-Meier database was used to analyze the impact of DHX9 expression on survival time.Then we used the immunohistochemistry to verify the difference of DHX9 expression in gastric cancer tissues and normal adjacent tissues,and uesd the western-blot experiment to verify expression of DHX9 in gastric cancer cells and normal gastric mucosal epithelial cells.After establishing of DHX9 stably silent expressed AGS and HGC-27 cells,we detected the effects of DHX9 on the proliferation,invasion,clonal formation ability and apoptosis of gastric cancer cells in vitro,such as CCK8,cloning,flow cytometry,scratch and Transwell test.Finally,we speculated the probable mechanism of DHX9 by analyzing the previous experimental results of our research group and consulting relevant literature.Results:1.The expression of DHX9 was high in both gastric cancer tissues and gastric cancer cells,which is closely related to the depth of tumor invasion and nerve invasion in gastric cancer patients.Meanwhile,patients with the high expression of DHX9 had shorter survival time and worse prognosis..2.After down-regulation of DHX9 expression,gastric cancer cells showed enhanced apoptosis,and clone formation,invasion and proliferation decreased..Conclusions:1.DHX9 is highly expressed in gastric cancer tissues and cells,and the high expression is significantly correlated with the depth of tumor invasion and nerve infiltration invasion in gastric cancer patients.2.The high expression of DHX9 shortens patient survival and is associated with poor prognosis.3.Down-regulation of DHX9 expression can inhibit proliferation,clonogenesis,invasion and promote apoptosis of gastric cancer cells. |
