Objective:To determine the expression level of long non-coding RNA HOST2 in gastric cancer,and to explore the clinical significance of lncRNA HOST2,and to study its effect on gastric cancer cells after interfering with lncRNA HOST2.Methods:1.Real-time fluorescence quantitative PCR was used to detect the expression of lncRNA HOST2 in gastric cancer and its corresponding paracancerous tissues,human gastric cancer cells and human gastric mucosal epithelial cells,and to study the correlation between the expression level and the clinicopathological data of patients.2.After interfering with lncRNA HOST2 in gastric cancer cells,the changes of proliferation,migration and invasion of gastric cancer cells were detected by CCK-8 cell proliferation,scratch test and Transwell invasion test.3.Western blotting(western blot)was used to detect the effect of lncRNA HOST2 on the expression of(EMT)-related proteins in gastric cancer cells.Results:1.The expression of lncRNA HOST2 in gastric cancer was significantly higher than that in paracancerous tissues,and the expression of lncRNA HOST2 was related to the depth of tumor invasion and lymph node metastasis.2.Compared with human gastric mucosal epithelial cells,the expression of lncRNA HOST2 was significantly increased in four human gastric cancer cell lines,and the highest expression level was found in MNK-45 cells.The interference efficiency was detected after interfering with lncRNA HOST2,among which Smur3 sequence had the best interference effect on gastric cancer cell lncRNA HOST2.3.After the interference of lncRNAHOST2 in gastric cancer cells,the ability of proliferation,migration and invasion of gastric cancer cells was significantly weakened.4.After the interference of lncRNAHOST2 in gastric cancer cells,the expression of EMT-associated protein E-cadherin(E-cadherin)was up-regulated and the expression of vimentin(Vimentin)was down-regulated.Conclusion:1.lncRNA HOST2 is highly expressed in gastric cancer tissues and gastric cancer cell lines,and is positively related to the depth of tumor invasion and lymphatic metastasis.2.lncRNA HOST2 can promote the proliferation,migration and invasion of gastric cancer cells by affecting the expression level of EMT-related proteins in gastric cancer cells.
