Ovarian cancer is the most common gynecological malignant tumor,and its pathogenesis is unknown.In the latest study,Ovarian Cancer is strongly associated with ferroptosis.The main mechanism of ferroptosis is the excessive accumulation of reactive oxygen species,destroys cell membranes and leads to deactivation of cells.Ferritin participates in the process of ferroptosis by regulating iron metabolism.In this experiment,the expression of FTL and FTH1 in normal ovarian tissue,benign ovarian tumors and carcinoma of ovary.The objective is to explore the process of ferroptosis in ovarian cancer and find a new treatment for ovarain cancer.Objective:To elucidate the expression of FTL and FTL in normal ovarian tissue,benign ovarian tumors and carcinoma of ovary.Methods:1.Immunohistochemistry was used to detect the expression of FTH1 and FTL in 32 cases of epithelial carcinoma of ovary,11 cases of benign ovarian tumor and 25 cases of normal ovarian tissue.2.Image processing:FTL and FTH1 expression were hign in the cytoplasm.FTL and FTH1 were positively expressed When the cytoplasm was dyed yellow or brown.The finished immunohistochemical sections were placed under the microscope to read the slices,and the visual field with good staining results was selected to take pictures.Each section was photographed 3 or more in different visual fields,and the pictures were uploaded to the computer system for preservation.Image analysis: the image analysis software image J was used to analyze the immunohistochemical staining intensity of FTL in epithelial ovarian cancer tissue,benign ovarian tumor tissue and normal ovarian tissue.Image J software will automatically calculate the Mean optical density(MOD)of each picture,and take the average of MOD acquired by Image J processing of the previous images of the same slice with different visual field,and the MOD of this section.3.Statistical analysis: SPSS24.0 was used for statistical analysis,and Shapiro-Wilk test was used to detect whether the dater accorded with the normal distribution.Student’s test was used to analyze the data conforming to normal distribution,and mean value±standard deviation was use to represent the result.Rank sum test was used to analyze the data did not accord with normal distribution,and median and percentile was use to represent the result.Pearson and Spearman correlation analysis method was used to analyze the correlation.P < 0.05 means there is statistical difference,P < 0.01 means there is significant statistical difference.Result:1.Expression of FTL protein in each groupThe median of MOD in FTL expression result in epithelial carcinoma of ovary,benign ovarian tumor and normal ovarian was 0.4273,0.1626 and0.2400,respectively.The expression of FTL was significantly different between epithelial carcinoma of ovary and benign ovarian tumor(P < 0.01),and the expression of FTL also was significantly different between epithelial ovarian cancer and normal ovarian(P < 0.01),but there was no statistical difference in FTL experssion between benign ovarian tumor and normal ovarian,(P > 0.05).It is suggested that the expression of FTL is significantly increased in epithelial carcinoma of ovary compared with benign ovarian tumor and normal ovarian.There was no statistical difference in the expression of FTL between patients with benign ovarian tumors and normal ovarian.2.Expression of FTL in epithelial carcinoma of ovary and analysis of its clinicopathological dataThe expression of FTL in epithelial carcinoma of ovary was not related to its FIGO stage(I-II stage,III-IV stage)and pelvic lymph node metastasis(p>0.05).There was no correlation between the expression of FTL and the tumor marker CA125 in epithelial carcinoma of ovary,and the Pearson correlation was 0.215(P > 0.05).There was no correlation between the expression of FTL and tumor marker HE4,age and p53 in epithelial carcinoma of ovary.The correlation coefficient of Spelman Rho was 0.076,0.021,0.181,respectively(P>0.05).3.Expression of FTH1 protein in each groupThe median of MOD in FTH1 expression result in epithelial carcinoma of ovary,benign ovarian tumor and normal ovarian was 0.5096,0.505 and0.4360,respectively.The expression of FTH1 wasn’t statistical different between epithelial carcinoma of ovary and benign ovarian tumor(P>0.05),but the expression of FTH1 was significantly different between epithelial ovarian cancer and normal ovarian(P < 0.01),there was no statistical difference in FTH1 expression between benign ovarian tumor and normal ovarian,(P > 0.05).It is suggested that the expression of FTH1 is significantly increased in epithelial ovarian cancer compared with normal ovarian.There was no statistical difference expression between of FTH1 protein in benign ovarian tumors and normal ovarian.And also there was no statistical difference expression between of FTH1 protein in benign ovarian tumors and epithelial ovarian cancers.4.Expression of FTH1 in epithelial carcinoma of ovary and analysis of its clinicopathological dataThe expression of FTH1 in epithelial carcinoma of ovary was not related to its FIGO stage(I-II stage,III-IV stage)and pelvic lymph node metastasis(p>0.05).There was no correlation between the expression of FTH1 and the tumor marker CA125 in epithelial ovarian cancer,and the Pearson correlation was-0.143(P > 0.05).There was no correlation between the expression of FTH1 and tumor marker HE4,age and p53 in epithelial ovarian cancer.The correlation coefficient of Spelman Rho was0.193,0.029,-0.211,respectively(P>0.05).5.Expression of FTL and FTH1 in normal ovarian tissueThere was a correlation between the expression of FTL and FTH1 in normal ovarian tissues,and the Pearson correlation was-0.563(P < 0.05).The expression of FTL was lower than that of FTH1,(P<0.01).The mean and median of MOD obtained by FTH1-FTL and FTH1/FTL were(0.1479,0.1884)(1.7475,1.7609),respectively.6.Expression of FTL and FTH1 in epithelial ovarian cancerThere was no correlation between the expression of FTL and FTH1 in epithelial ovarian cancer,and the Pearson correlation was-0.160(P >0.05).However,the overall expression level of FTH1 in the sample was higher than that of FTL,(P<0.05).The mean and median of MOD obtained by FTH1-FTL and FTH1/FTL were(0.0901,0.1110)(1.4525,1.2916),respectively.Conclusions1.FTL and FTH1 subunits of ferroptosis associated protein-ferritin are highly expressed in epithelial ovarian cancer,suggesting that FTL and FTH1 may promote the occurrence and development of ovarian cancer.2.In the process of ovarian cell carcinoma,the correction between FTL and FTH1 of two subunits of ferritin is lost.And the ratio of FTH1 to FTL is decreased,which further suggests that FTL and FTH1 may be related to the occurrence of ovarian cancer.3.FTL and FTH1 may be a new target for the treatment of ovarian cancer.
