"Zang Yinchen" is a famous Tibetan medicine for the treatment of hepatobiliary diseases in Tibet,Qinghai,Sichuan,Yunnan,Gansu and other regions of China.There are more than 40 species of original plants,including Swertia,Gentiana,Gentianopsis,Halenia,and Lomatogonium in Gentianaceae and Saxifraga in Saxifragaceae.The main representative medicinal material is Swertia mussotii Franch.It is the authentic medicine in many medicines.However,due to the limited wild resources of S.mussotii.,Halenia elliptica D.Don and Gentianopsis paludosa(Hook.f.)Ma are the most commonly used substitutes in pharmaceutical production.Therefore,in this study,S.mussotii.,H.elliptica.and G.paludosa.were taken as the research objects,and the chemical composition,in vitro anti-inflammatory activity and molecular simulation mechanism of the above three medicinal materials were compared.Try to explore the material basis and theoretical basis of these three medicinal materials through comparative study.The main results of this paper are as follows :1.Through Box-Behnken analysis in response surface,the optimum extraction process of iridoid glycosides from S.mussotii.was established,which improved the content of iridoid glycosides in active components and the utilization rate of medicinal materials.2.The main chemical components of the three Tibetan medicinal materials Zang Yinchen were analyzed by LC-MS.Through structural analysis and comparative study,it was found that the main structural types of the three kinds of medicinal materials were basically the same,including iridoid glycosides,xanthones and flavonoids.However,the same components in the main chemical components of the three are very few,so it can not be proved that the three medicinal materials can be mixed use from the perspective of chemical composition.3.The in vitro anti-inflammatory activity of crude extracts of three kinds of Zang Yinchen medicinal materials was studied by RAW-264.7 cell inflammation model in vitro.It was confirmed that the three kinds of crude extracts could significantly inhibit the production of NO,TNF-α and IL-1β and promote the expression of IL-10 in cells at the concentration of 40 μg/m L.By comparison,it was found that although there were some differences in the effects of the three extracts on the expression of inflammatory factors,the overall anti-inflammatory activities in vitro were basically the same.4.Based on network pharmacology,molecular docking and molecular dynamics,it was confirmed that more than 12 active compounds in the three herbs acted on the core targets such as SRC and MAPK1 and 15 common signal pathways such as PI3 KAkt,MAPK1,JAK-STAT and AGE-RAGE to exert their anti-hepatitis and other hepatic diseases.It shows that the three kinds of Tibetan medicinal plant Zang Yinchen are also highly consistent in the regulation of disease pathways.Through the comparative study of chemical composition,in vitro activity and molecular mechanism,it was found that although there were great differences in the main chemical components of the three original Tibetan medicinal plant Zang Yinchen,their in vitro activity and anti-inflammatory mechanism were consistent,which may be the basis for their substitution.However,to fully clarify the rationality of its substitution,a large number of in vivo model validations are still needed.
