The Relationship Between CACNA2D2 And Clinical Stage And Prognosis Of Lung Adenocarcinoma

Background and purpose:In recent years,many molecular markers have been found to be associated with the growth,proliferation,metastasis,and prognosis of lung adenocarcinoma.Although these molecular markers have been proven to play an important role in the prognosis evaluation and treatment strategy formulation of lung adenocarcinoma.However,there are still significant limitations in many aspects,such as high cost of genetic testing or significant trauma during the testing process.Therefore,developing molecular markers related to lung adenocarcinoma that are easy to detect and minimally invasive during the sampling process is still of great significance for the early diagnosis,treatment,and prognosis evaluation of lung adenocarcinoma patients.Method:1.The collection and organization of the database obtained the TCGA-LUAD dataset from the official website of TCGA,which includes the predicted gene set for the expression profiles of cancer tissue and normal tissue samples from 585 LUAD patients.By searching the GEO database,the GSE19188 dataset,GSE20189 dataset,and GSE27554 dataset can be obtained.The GSE19188 dataset includes a predictive gene set for the expression profiles of 91 lung cancer and 65 adjacent normal lung tissue samples.The GSE20189 dataset includes a gene expression profile of peripheral whole blood(PWB)from 73 adenocarcinoma cases and 80 control group peripheral whole blood(PWB).The GSE27554 dataset includes a gene expression profile of normal patient bronchial lining fluid and lung cancer patient bronchial lining fluid,Select the common differentially expressed genes from the results of these four sets of differential analysis.Then,the biological function of the gene was studied through GO and KEGG enrichment analysis.Comparative analysis of clinical pathological features and key gene expression was conducted using Wilcoxon signed rank test and Kruskal Wallis test.Finally,the Kaplan Meier method was used to perform survival analysis on the LUAD patient dataset in the public database.2.The human protein atlas(THEHUMANPROTEINATLAS,https://www.proteinatlas.org)is an online database.The data comes from the whole gene protein sequences obtained after the genome sequencing of various species is completed.This database collects immunohistochemical staining images of LUAD and normal tissues,and through this database,the expression differences of CACNA2D2 in LUAD and normal tissues can be verified.Results:1.Through bioinformatics analysis,we found that the expression level of CACNA2D2 in lung adenocarcinoma tissue is lower than that in normal adjacent tissues;Subsequently,the GSE19188,GSE20189,and GSE27554 datasets were analyzed using R,and the results showed that the expression levels of CACNA2D2 in LUAD tissue,peripheral blood of LUAD patients,and bronchial lining fluid of LUAD patients were lower than those in adjacent cancer tissue,control group peripheral blood,and control group bronchial lining fluid,respectively.2.The results of immunohistochemistry in human protein profile data indicate that the expression level of CACNA2D2 is significantly increased in normal lung tissue,while there is a significant decrease in CACNA2D2 expression in lung adenocarcinoma tissue.Conclusion:Through the analysis of sample data in TCGA and GEO,combined with the immunohistochemical results of CACNA2D2,it was found that the expression of CACNA2D2 was downregulated in LUAD tissue,and the expression levels of CACNA2D2 in bronchial lavage fluid and peripheral blood in the LUAD group were lower than those in the non LUAD group.The results indicate that low expression of CACNA2D2 may be an independent prognostic factor for poor survival in LUAD.
