| Background:Brain glioma is the most common malignant tumor of the central nervous system,among which glioblastoma is the most malignant type,which is highly invasive and heterogeneous,accounting for 2/3 of all diagnoses of diffuse glioma,the median survival period is only 12-15 months,and the current treatment methods are still unable to control the 5-year progression-free survival and overall survival of tumor patients,especially advanced patients.Glioma patients often face tumor relapse、progression and malignant transformation after comprehensive treatment.Recent studies have found that there is a certain proportion of tumor stem cells in gliomas,which participate in the formation of gliomas,generate tumor heterogeneity,and are inseparable from the generation of therapeutic resistance of gliomas.Studies have found that Bmi1 can be used as one of the marker factors of tumor stem cells.Gli1 is a key signaling factor in the Hedgehog signaling pathway.The aberrant activation of Gli1 is related to tumor proliferation,invasion and tumor stemness,but the expression and clinical significance of the two in glioma are still unclear.Objective:The purpose of this project is to study the expression of Bmi1 and Gli1 in brain glioma,explore the associations between Bmi1 and Gli1 expression with the clinicopathological characteristics and prognosis of patients with glioma,and preliminarily study the relationship between Bmi1 and Gli1.Whether there is a connection between them will lay the foundation for the subsequent research on the molecular mechanism of glioma.Methods:1.By selecting 86 patients from the Second Affiliated Hospital of Nanchang University who were pathologically diagnosed with glioma from January 2017 to December 2019,the complete medical records and clinical information were collected,and the overall survival time of the patients was followed up.2.Screen the appropriate wax blocks for immunohistochemical staining using the En Vision two-step method,and score semi-quantitatively under a microscope.3.Analyze the relationship between the expression of Bmi1 protein and Gli1 protein and the clinicopathological parameters of glioma patients,and analyze the impact of the expression of the two proteins on the overall survival of glioma patients Results:1.The expression of Bmi1 protein and Gli1 protein in the brain tissue of patients with cerebrovascular malformation and glioma of different grades was statistically different(p<0.001).2.The expression of Bmi1 protein in glioma is related to histological grade(p<0.05),and is not related to patients’ age,sex,tumor growth site,and maximum tumor diameter,and postoperative radiotherapy and chemotherapy(p>0.05).The expression of Gli1 protein in glioma was related to the histological grade of glioma(p<0.05).It was not related to patients’ age,sex,tumor growth site,maximum tumor diameter,whether postoperative radiotherapy and chemotherapy,(p>0.05).3.The expression of Bmi1 and Gli1 protein was positively correlated with the pathological grade of glioma,and the patients with high expression had higher tumor grade(p<0.05).4.Univariate survival analysis showed that patients’ age,gender,tumor histological grade,Bmi1,and Gli1 protein expression were associated with poor prognosis of glioma patients(p<0.05),multivariate survival analysis showed that patients’ age,tumor histological grade,and Bmi1 protein expression could be independent prognostic factors for glioma patients(p<0.05).Conclusion:1.The expression of Bmi1 and Gli1 in glioma is related to tumor grade,and the expression in high-grade glioma is significantly higher than that in normal brain tissue and low-grade glioma.There is a potential relationship between the expression of Bmi1 and Gli1 in glioma at the histological level.2.The high expression of Bmi1 and Gli1 in glioma is related to the malignant progression and poor prognosis of the tumor,which can be used as potential therapeutic targets for glioma patients. |
