| Purpose:Cervical cancer is the most common malignant tumor of the female reproductive system.The pathological types are mainly divided into squamous cell carcinoma and adenocarcinoma,of which cervical squamous cell carcinoma accounts for 80%,and cervical adenocarcinoma accounts for 5-20%.The treatment effect and prognosis are also different.Therefore,we analyzed the similarities and differences between cervical cancer tissues and cells,and on this basis,we identified key genes that may be associated with the progression of cervical squamous cell carcinoma and adenocarcinoma to find potential markers or therapeutic targets.Methods:Obtain the gene expression profile data and clinical data of cervical cancer tissues and normal cervical tissues from TGCA and GTEx databases from the UCSC platform,and determine the differentially expressed genes(DEGs)between cancer tissues of patients with cervical squamous cell carcinoma and adenocarcinoma and normal cervical tissues.The construction and sequencing of the RNA-Seq library of cervical squamous cell carcinoma Siha and adenocarcinoma He La cell lines were carried out by Beijing Nuowei Gene Technology Co.,Ltd.(Beijing,China).Then,the potential functions of DEGs were analyzed through the DAVID bioinformatics database.The STRING online website constructs a protein-protein interaction(PPI)network,Cytoscape software(version 3.9.1)performs visual analysis of the PPI network,and the Hub plug-in(version 2.0.0)is used to identify the central genes of the PPI network.The correlation between the expression levels of central genes and the prognosis of patients with cervical squamous cell carcinoma and adenocarcinoma was analyzed by R software,and the association between survival-related gene expression and immune infiltration was explored by TIMER2.0.Results:In this study,we found 1398 DEGs in cervical squamous cell carcinoma and adenocarcinoma,among which 536 DEGs were significantly up-regulated and 662 DEGs were significantly down-regulated in squamous cell carcinoma compared with adenocarcinoma;compared with He La cells,There were 6869 DEGs in Siha cells,among which 3221 DEGs were relatively significantly upregulated and 3648 DGEs were relatively significantly downregulated.Bioinformatics analysis shows that there are some differences in molecular expression levels and biological processes between different types of cervical cancer cell lines and clinical tissues.Based on this,we analyzed the common DEGs between the two.Through PPI network analysis,we determined that There are 10 central genes in squamous cell carcinoma and adenocarcinoma respectively.The central genes of squamous cell carcinoma include EGFR,ERBB2,IL1 B,MMP9,NOTCH1;the central genes of adenocarcinoma include IL1 B,MMP9,EPCAM,SOX9,and CAV1.Survival analysis showed that the expression of ERBB2 and EGFR was negatively correlated with the prognosis of patients with cervical adenocarcinoma;MMP9 was negatively correlated with the prognosis of patients with cervical squamous cell carcinoma,and SOX9,CAW1,and IL-1B were positively correlated.In addition,the relationship between prognosis-related genes and immune infiltration was analyzed.Conclusions:In this study,we identified DEGs between cervical squamous cell carcinoma and adenocarcinoma,as well as the similarities and differences between tissues and cells.In addition,we identified 10 hub genes in each of squamous cell carcinoma and adenocarcinoma.This study may provide new biomarkers for the prognosis of squamous cell carcinoma and adenocarcinoma. |
