| Background and Objective: The prognosis of patients with lung squamous cell carcinoma(LUSC)is poor.The role of mi RNAs in the occurrence,development and diagnosis of LUSC has gradually attracted the attention of scholars.In order to further enrich the understanding of mi RNAs in the diagnosis and treatment of LUSC,explore the key mi RNAs related to the occurrence and development of LUSC,and their impact on the prognosis of patients,we used the Bioinformatics Method to study and analyze the expression of mi RNAs in LUSC.Methods: The expression profile data downloaded from the Cancer Genome Atlas(TCGA)database were analyzed by R language integration,and differential expression genes(DEG)were screened from it;the data set was screened in the Gene Expression Omnibus(GEO)database as validation according to the inclusion criteria;the survival analysis of DEG was carried out by SPSS software;with the help of DAVID database,GO annotation and KEGG enrichment analysis are carried out for DEG,and the interaction path map of enrichment function is drawn.Results:(1)a total of 52 DEGs were screened from TCGA database,38 of which were up-regulated and 14 of which were down-regulated;(2)four up-regulated mi RNAs and one down-regulated mi RNA were finally obtained through the verification of GEO database;(3)survival analysis showed that the expression level of mi R-210 was significantly related to the overall survival(OS)of patients,and further analysis showed that in the up-regulated expression group of mi R-210,the OS of patients without radiotherapy was significantly better than that of patients with radiotherapy;(4)the function of mi R-210 was concentrated,and the enrichment function of mi R-210 was mainly concentrated in the positive and negative cell proliferation regulation,transcription of DNA and metabolism of choline;the signal pathway mainly focuses on vascular endothelial growth factor(VEGF)signal pathway,hypoxia inducible factor-I(HIF-1)signal pathway,mitogen activated protein kinases(MAPK)signal pathway,etc.Conclusion: Mir-210 is an independent prognostic factor in patients with LUSC.The OS is not only related to T stage of tumor,but also to the expression of mir-210.In addition,in the up-regulated expression group of mir-210,the patients had poor response to radiotherapy.Combined with the relationship between HIF-1 and radiation tolerance,the up-regulated expression state of mir-210 was more likely to produce radiotherapy resistance,which suggests that mir-210 may become an important reference for clinical treatment decision-making of LUSC patients in the future. |
