| Objective:To explore the mechanism of ethanol extract of Euonymus alatus on CCl4-induced hepatic fibrosis in mice by regulating JAK2/STAT3signaling pathway.Methods:Sixty C57BL/6J mice were randomly divided into control group,model group,EAL,EAM,EAH,and Silybin group(n=10).Except for the control group,mice in other groups were injected with 25%CCl4of 1.6m L·kg-1to induce HF model.Moreover,EAL,EAM,and EAH were administered72,140 and 280 mg·kg-1ethanol extract of euonymus alatus once by gavage once a day,and the positive group were administered 12.6 mg·kg-1Silybin by gavage once a day,and the intervention lasted 6 weeks.After 6th week,mice blood was collected,the body weight and liver weight were massured,the liver appearance was observed,the liver mass index was calculated,and the serum ALT,AST,TNF-α,IL-6,IL-1βlevel were detected.The protein expression levels of COL-1,α-SMA,and JAK2/STAT3 signal pathway-related protein(JAK2,STAT3,p-JAK2,p-STAT3)in mice liver tissues were detected.Results:Compared with the blank group,the liver index of the model group was significantly increased.The liver cell structure was disordered,and the liver cell nucleus was displaced,disappeared or enlarged.Produce false lobules,inflammatory infiltration,fibrous tissue hyperplasia,and collagen aggregation in the liver tissue space.The serum expression levels of ALT,AST,TNF-α,IL-6,and IL-1βwere signific-antly increased(P<0.05).The protein expression levels ofα-SMA,COL-1,JAK2,STAT3,p-JAK2,p-STAT-3 were significantly increased in liver tissues(P<0.05).Compared with the model group,the liver index had significantly decreased(P<0.05),the liver morphology,inflammatory infiltration,fiber changes,and collagen deposition had improved in EAL,EAM and EAH groups mice liver tissues.The serum expression levels of ALT,AST,TNF-α,IL-6,IL-1βwere significantly decreased(P<0.05).And the protein expression levels ofα-SMA,COL-1,JAK2,STA-T3,p-JAK2,p-STAT3 were significantly increased in liver tissues(P<0.05).Conclusion:Ethanol extract of Euonymus alatus have an anti CCl4-induced HF effect in mice,and its mechanism may be related to the regulation of the JAK2/STAT3 signaling pathway. |
