| Objective : Sarcopenia in elderly women has become an important problem affecting the quality of life of the elderly in the context of global aging.Studies have confirmed that resistance exercise,aerobic exercise and estrogen replacement therapy have positive effects on skeletal muscle health,but as a passive exercise method with a lower threshold,whole-body vibration training deserves more exploration.At present,there are few reports on whether whole body vibration training combined with estradiol supplementation can effectively improve skeletal muscle mass and enhance skeletal muscle function.This study intends to construct an animal model of sarcopenia through ovariectomy,to explore the effects of whole-body vibration training combined with estradiol supplementation on skeletal muscle mass,function and the expression of related signaling proteins,so as to improve the treatment of whole-body vibration training combined with estradiol supplementation Elderly female sarcopenia provides experimental and theoretical basis.Methods : 40 female C57 mice were selected as experimental research objects,and the mice were adaptively reared for 2 weeks and then underwent ovariectomy(OVX)to establish a female mouse model of sarcopenia.After successful modeling,32 of the remaining 36 animals(4lost due to postoperative diseases and other reasons)were selected and randomly divided into 4 groups with 8 animals in each group.Divided into ovariectomized group(OVX),ovariectomized estradiol supplementation group(OVX+E),ovariectomized whole body vibration training group(OVX+W),ovariectomized whole body vibration training plus estradiol supplementation group(OVX+W+ E).Then the mice in the OVX+E group and the OVX+W+E group were operated,and the subcutaneous tissue was embedded with 17β-estradiol sustained-release tablets,0.36 mg/tablet;The rats were subjected to whole-body vibration training.First,adaptive vibration training was performed for 3 days,once a day,for 15 minutes each time.After the adaptive training,whole body vibration training intervention was carried out for 8 weeks,5 days a week,once a day,30 min each time,the vibration frequency was 30 Hz,and the amplitude was 3 mm.The OVX group and the OVX+E group were fed normally.After the intervention,body composition and grip strength were measured;bilateral quadriceps muscles were weighed;serum estradiol level was detected by Elisa method;P-AKT/AKT,P-Fox O1/Fox O1,and P-m TOR were detected by Western blotting technique /m TOR protein expression level.Results:1.After the experiment,there was no significant difference in the body weight of mice in each group;the lean body mass and the ratio of lean body mass in the OVX+E group and the OVX+W group were significantly higher than those in the OVX group(p<0.05);at the same time,the OVX+W+E group The lean body mass and proportion of lean body mass were significantly higher than those in the OVX+W group(p < 0.05).2.The detection results of serum estradiol showed that the serum estradiol levels in the OVX+E group,the OVX+W group and the OVX group were significantly increased(P<0.05).Diol levels were significantly increased(P<0.05).3.The results of grasping force of mice showed that the grasping force of mice in OVX+W group and OVX+W+E group increased significantly after experimental intervention compared with before experimental intervention(P<0.05);at the same time,after experimental intervention,OVX+Compared with the OVX+W group,the grasping force of the mice in the W+E group was significantly increased(P<0.05).4.The bilateral quadriceps femoris were weighed after sampling.The results showed that compared with the OVX group,the quadriceps muscle mass of the mice in the OVX+W group and the OVX+E group was significantly increased(P<0.05).Compared with the OVX+W group,the quadriceps muscle mass of the mice in the OVX+W+E group was significantly increased(P<0.05).5.The results of Western Blot showed that compared with the OVX group,the ratios of P-AKT/AKT and P-m TOR/m TOR in the OVX+W group and OVX+E group were significantly increased(P<0.05);P-FoxO1/FoxO1 The ratio decreased significantly(P<0.05).At the same time,the ratios of P-AKT/AKT and P-m TOR/m TOR were also significantly increased in the OVX+W+E group compared with those in the OVX+W group(P<0.05),and the ratio of P-Fox O1/Fox O1 was significantly decreased(P<0.05).).6.The ratio of P-AKT/AKT and P-mTOR/mTOR was positively correlated with the level of serum estradiol;the ratio of P-Fox O1/Fox O1 was negatively correlated with the level of serum estradiol.Conclusions:(1)Whole body vibration training combined with estradiol supplementation significantly improved skeletal muscle mass and optimized body composition in ovariectomized mice.(2)Whole body vibration training can effectively improve the muscle strength of ovariectomized mice and increase the serum estradiol level in ovariectomized mice.(3)The increase of serum estradiol level can effectively promote the synthesis of skeletal muscle and inhibit the decomposition of skeletal muscle,and its possible mechanism is related to the expression of related signaling proteins. |
