Objective: To explore the difference of serum proteomics between cervical spondylotic myelopathy(Qi and blood stasis syndrome)and healthy people,and to search for and identify the potential biological markers of CSM(Qi and blood stasis syndrome).Methods: Serum samples were collected from 30 patients with cervical spondylotic myelopathy(CSM-Q)and 30 healthy subjects(HC).Comparative isotope labeling and absolute quantification(TMT)combined with liquid chromatography-tandem mass spectrometry(LC-MS/MS)was used for proteomic analysis to find and identify differentially expressed proteins.Uni Prot GOA was used to generate gene ontology(GO)annotation,and Kyoto Encyclopedia of genes and genomes(KEGG)database was used to analyze the enrichment pathway The protein-protein interaction network(PPI)was constructed using the STRING database(v.10.5).Results:(1)A total of 78 distinct proteins were screened by TMT,of which 12 were up-regulated and 66 were down-regulated.(2)Based on GO pathway enrichment analysis,differential proteins are involved in protein kinase B binding,angiostatin binding,immunoglobulin receptor binding,proton transmembrane transporter activity,translation elongation factor activity,ferrous binding,active ion transmembrane transporter activity,antigen binding,and active transmembrane transporter activity.(3)Based on the enrichment analysis of KEGG pathway,7 common differential signal/metabolic pathways were screened out,signal pathways include mineral absorption,oxidative phosphorylation,platelet activation,neutrophil trap formation,and Huntington’s disease.(4)PPI analysis showed that 21 common differential proteins(IGLC2,IGLC7,IGLL5,IGLV2-8,IGLV3-25,IGKV3D-11,ITGA2 B,FGA,FGG,FGB and ALB)were located at the nodes of the protein-protein interaction network,it is closely related to biological processes such as immune reaction,inflammatory reaction,coagulation reaction and oxidative stress reaction.Conclusion: TMT combined with LC-MS/MS technique was successfully used to screen the differential expression proteins between CSM(Qi and blood stasis syndrome)and healthy people,it is clear that immunoglobulin,integrin,fibrinogen and endoplasmic reticulum calcium-binding protein may be the specific serum markers of cervical spondylotic myelopathy(Qi and blood stasis syndrome).
